dc.description.abstractSão poucos os criatórios industriais de pato doméstico no Brasil, pois para a população nacional, carne e ovos desta ave ainda são considerados restritos. As criações de subsistência são comuns no país e o comércio de aves vivas, abatidas, ovos e carne ocorrem principalmente entre os pequenos produtores, casas comerciais e feiras livres. Não existe registros ou conhecimento das condições higiênico sanitárias onde essas aves são criadas. A classe das aves está suscetível a se infectarem por três espécies de Cryptosporidium (Cryptosporidium meleagridis, C. baileyi e C. galli), além destas 13 genótipos. O trabalho teve como objetivos: diagnosticar microscopicamente a presença de Cryptosporidium spp. no pato doméstico da família Anatidae (Cairina moschata), provenientes de duas criações rústicas localizadas no Rio de Janeiro; realizar a caracterização genotípica seguida do sequenciamento e análises filogenéticas, comparando com as sequencias obtidas com aquelas sequências depositadas no GenBank. Como metodologia, amostras fecais de patos domésticos (Cairina moschata) foram colhidas em duas criações rústicas (Área A e B), totalizando 118 amostras, sendo 60 amostras da área A e 58 da área B. Os animais da coleta eram aves jovens que não ultrapassaram 30 dias de vida. Inicialmente foi realizada a pesquisa de oocistos Cryptosporidium spp., através da técnica de centrifugação e flutuação em solução saturada de açúcar. No diagnóstico microscópico, de um total de 118 amostras analisadas 20 apresentaram positividade para Cryptosporidium (118/16,95%), sendo estas pertencentes a área A (60/ 33,33%). Para a realização da extração de DNA, foram selecionadas aquelas que apresentavam mais de 20 oocistos de Cryptosporidium spp. por campo microscópico, seguida de metodologias para o diagnostico molecular tendo como alvo o 18S, que posteriormente foram sequenciadas e análises filogenéticas foram feitas. dc.description.abstractOtherThere are few industrial duck farms in Brazil, because for the national population, the meat and eggs of this bird are still considered restricted. Subsistence creations are common in the country, and trade in live, slaughtered poultry, eggs and meat mainly occurs among small producers, commercial houses and free markets. There are no records or knowledge of the hygienic sanitary conditions where these birds are created. The class of birds is susceptible to infection by three species of Cryptosporidium (Cryptosporidium meleagridis, C. baileyi and C. galli), in addition to these 13 genotypes. The objective of this study was to diagnose the presence of Cryptosporidium spp. in the domestic duck of the family Anatidae (Cairina moschata), from two rustic creations located in Rio de Janeiro; Carry out the genotypic characterization followed by sequencing and phylogenetic analyzes, comparing with the sequences obtained with those sequences deposited in GenBank. As a methodology, fecal samples of domestic ducks (Cairina moschata) were collected in two rustic creations (Area A and B), totaling 118 samples, 60 samples from area A and 58 from area B. The animals of the collection were young birds that did not exceed 30 days of life. Initially the research of oocysts Cryptosporidium spp. was carried out, through the technique of centrifugation and flotation in saturated solution of sugar. In the microscopic diagnosis, 20 samples from area A showed positivity (118 / 16.95%). Of these, we selected those that had more than 20 oocysts of Cryptosporidium spp. By microscopic field for the extraction of DNA, followed by molecular diagnostic methodologies targeting 18S, which were later, sequenced and phylogenetic analyzes were performed.
