dc.description.abstractLesões cardíacas agudas ou crônicas podem resultar em perturbações na função renal e vice versa, fenômeno conhecido como Síndrome Cardiorrenal. Este trabalho teve como objetivos verificar, com base na morfologia, as correlações entre Doença Renal Crônica (DRC) e lesões cardiovasculares, avaliar a real extensão dessas lesões em cães, visto que alterações miocárdicas regressivas incipientes podem não ser perceptíveis ou facilmente identificadas através da coloração rotineira pela hematoxilina e eosina, bem como verificar a eficácia do teste imunohistoquímico com o uso do anticorpo antitroponina C humano na detecção de lesões cardíacas em cães com insuficiência renal. Pela avaliação imunohistoquímica do coração dos 22 caninos, observaram-se diversos grupos de miócitos com redução significativa ou ausência de imunorreatividade para o anticorpo anti-troponina C, essas áreas, via de regra, correspondiam aos mesmos grupos de miócitos que, pela coloração de H.E. apresentavam alterações que variavam de leve tumefação celular a aumento da eosinofilia, perda de estriação, lise celular e cariólise, por vezes, acompanhadas de infiltrado inflamatório predominantemente mononuclear. Adicionalmente, havia, no coração de 10 cães, lesão vascular, que variou de leve tumefação das células endoteliais a necrose, com deposição de material eosinofílico (marcação positiva para colágeno pela coloração Tricrômico de Masson), na parede vascular e mineralização. Macroscopicamente, observaram-se hipertrofia ventricular esquerda e lesões extra-renais da uremia, incluindo glossite e estomatite ulcerativas, pneumopatia e gastropatia urêmica, além de mineralização do espaço intercostal. Macroscopicamente os rins avaliados apresentaram atrofia, superfície irregular, fibrose e alteração na relação córtico-medular e na microscopia hialinização e esclerose glomerular, necrose marcada nas células tubulares, mineralização de algumas estruturas, inflamação (predominantemente mononuclear) e fibrose intersticial. Neste estudo, as lesões miocárdicas foram estreitamente correlacionadas às alterações vasculares, resultantes da uremia de longa duração e/ou da combinação de alterações metabólicas e celulares que ocorrem na Síndrome Cardiorrenal tipo 4, na qual o comprometimento crônico do rim pode induzir lesões crônicas no coração.por

dc.description.abstractOtherAcute or chronic abnormalities in cardiac function can lead to lesions in kidneys or heart and cause the Cardiorenal syndrome. This study had the objective to (1) to verify, based on morphology, the correlation between Chronic Kidney Disease (CKD) and cardiovascular injury, as well as to reveal the extent of the lesion in dogs; because initial regressive changes often cannot be discerned with hematoxylin and eosin staining (H&E) alone, and (2) to verify the reliability of immunohistochemistry with anti-cTnC human antibody, to detect heart lesions in dogs with renal failure. Cardiac fragments from 22 dogs were submitted to immunohistochemistry, which revealed reduction of cTnC levels in the cytoplasm of cardiomyocytes. This decrease in immunoreactivity occurred usually on the same myocytes with specifics changes in H&E stain, ranging from mild cell swelling to increased eosinophilia, loss of striation, cell lysis and karyolysis that sometimes was accompanied by a inflammatory infiltrate. In 10 cardiac fragments vascular injury was observed, ranging from mild swelling to necrosis of endothelial cells, with deposition of eosinophilic material (staining positively for collagen in Masson’s trichrome) in the vessel wall and mineralization. Grossly, there were left ventricular hypertrophy and extra-renal lesions of uremia, including glossitis and ulcerative stomatitis, uremic pneumo and gastropathy, and intercostal mineralization. The evaluated kidneys had lesions of differtnmet intensity, irregular surface, fibrosis and altered cortico-medullary relationship, hyalinization and glomerular sclerosis, marked tubular cell necrosis and mineralization, mononuclear infiltration, and interstitial fibrosis. In this study, the cardiac vascular lesions were correlated with changes resulting from long term uremia and/or combination of metabolic and cellular changes of the Cardiorenal syndrome type 4, wherein the chronic kidney impairment can induce chronic lesions in the hearteng
