dc.description.abstractEste estudo teve por objetivo caracterizar a diversidade gênica de estirpes de Azospirillum amazonense isoladas do solo da rizosfera, do rizoplano e do interior radicular dos cereais arroz (C3), milho e sorgo (C4), cultivados em dois solos distintos, ltaguaí e Ecologia, procurando encontrar evidências de um eventual efeito discriminatório exercido pelo solo e/ou planta. A estratégia de investigação baseou-se (i) em experimentos de hibridização dot-blot com sondas oligonucleotídicas 23S DNAr grupo-, gênero- e espécie- específicas e (ii) nas análises de restrição dos fragmentos intergênicos 16S-23S DNAr amplificedos. Os resultados gerados pelas 7 enzimas de restrição foram posteriormente analisados através das metodologias de agrupamento e de análise da variância molecular (AMOVA). Somente 28 % dos isolados, em sua maioria do solo Ecologia, foram capazes de hibridizar com a sonda específica para a espécie A. amazonense, AA 23S. O padrão de hibridização dos isolados pode estar parcialmente associado ao tipo de solo. Os resultados gerados pelas análises de agrupamento e pela AMOVA revelaram a existência de 4 grupos geneticamente distintos, discretos e intemamente heterogêneos. Os níveis de diversidade encontrados em cada grupo foram elevados. Não foi observada nenhuma diferença significativa na estrutura, nos padrões genotípicos e nos níveis de diversidade entre os isolados do solo da rizosfera + rizoplano e os do interior radicular. O tipo de solo/ambiente exerce aparentemente um efeito discriminatório sobre os isolados de A. amazonense, em especial sobre os isolados da rizosfera e do rizoplano. A planta, por sua vez, é capaz de, até certo limite, influenciar a diversidade populacional de A. xiii amazonense. Plantas de arroz cultivadas no solo ltaguaí e, menos intensamente, plantas de sorgo cultivadas no solo Ecologia discriminaram parcialmente os isolados do solo da rizosfera e das raízes lavadas. Plantas de arroz, milho e, menos intensamente, sorgo, cultivadas no solo ltaguaí, exerceram efeitos discriminatórios sobre os isolados do interior radicular. Os resultados e conclusões gerados neste estudo evidenciam de maneira clara a conveniência de se utilizar de maneira complementar as análises de agrupamento e da variância molecular para a caracterização de populações de bactérias e a determinação dos efeitos discriminatórios exercidos pelo solo e a planta.pt_BR

dc.description.abstractOtherIn this study, the populational structure of Azospirillum amazonense isolates recovered from the rhizosphere soil, washed and sterilized roots of rice (C3), maize and sorghum (C4) plants, cropped in two distinct soils, Itaguaí and Ecologia, was characterized. Discriminatory effects exerted by the soil and plant types were also investigated. Two methodological approaches based on (i) dot-blot hybridization experiments with group, gender- and species-specific 23S rDNA based oligonucleotide probes and (ii) restriction fragment length polymorphism analysis of the 16S-23S rDNA intergenic spacer region, followed by proper statystical analysis (cluster analysis and analysis of molecular variance- AMOVA), were used. A. amazonense isolates showed a characteristic hybridization pattern, since only 28% of the isolates hybridized with the A. amazonense-specific probe AA 23S. This hybridization pattern seems to be associated with the soil type. Four geneticelly distinct and highly heterogeneous clusters formed at 78% similarity were observed and diversity level within each cluster was very high. There were no differences in structure, genetical pattems and diversity levels between isolates recovered from rhizosphere + rhizoplane and from the inside of the roots. Aparently the soil/environment type is able to discriminate A. amazonense Mates, specially those recovered from rhizosphere soil and rhizoplane. The plant type also seems to exert a discriminatory effect. Rice and sorghum plants, cropped in ltaguaí and Ecologia soils, respectively, partially discriminated the thizosphere and rhizoplane isolates. Rice, maire and sorghum plants cultivated in Itaguaí soil were able to discriminate the endophytic isolates. The results and conclusions generated in this study clearly demonstrated the convenience of utilising cluster analysis and AMOVA for characterization of bacterial populations and determination of discriminatory effects exerted by soil and plant types.en
