| dc.description.abstract | A produção de hidrogênio durante a redução enzimática do N2 para a amônia é um dos fatores que mais influenciam a eficiência da fixação biológica do nitrogênio. Em soja, a eficiência relativa da fixação de nitrogênio tem sido relacionada a abundância relativa de ureídos no exsudato xilemático e a partição do nitrogênio para as sementes. Tendo em vista a importância econômica e ecológica da utilização da FBN em soja, tem-se buscado estirpes que proporcionem um aumento na produção de grãos da cultura. Além disso, esforços tem sido feitos visando conhecer os mecanismos e fatores que afetam o sistema simbiótico. A influência da hidrogenase na fixação de N2, no transporte e na partição do nitrogênio foi avaliada em plantas noduladas de soja. No primeiro experimento, avaliou-se o efeito da inoculação da soja com as estirpes CB 1809 (Hup+) de Bradyrhizobium japonicum e 29W (Hup-) de B. elkanii na FBN e na partição do nitrogênio na parte aérea. A FBN foi estimada pelo método de diluição isotópica de 15N. Cerca de 95% do nitrogênio total da parte aérea foi proveniente da fixação de N2. A estirpe Hup+ fixou mais N-total do que a estirpe Hup-. A atividade dos nódulos formados pela estirpe Hup- foi drasticamente reduzida durante o período de formação de vagens, enquanto, a estirpe Hup+manteve elevados níveis de fixação de N2 durante todo o período reprodutivo da soja. A maior atividade dos nódulos formados pela estirpe Hup+ refletiu em um aumento na proporção de N-ureídos no exsudato xilemático das plantas durante o período reprodutivo. As plantas inoculadas com a estirpe CB 1809 apresentaram um maior índice de colheita de biomassa e nitrogênio e uma maior produção de grãos em comparação com a estirpe 29W. Na fase sequinte, plantas de soja inoculadas com as estirpes CB 1809 (Hup+) ou 29W (Hup-) foram cultivadas sob luminosidade total ou sombreadas (70% de sombreamento), e amostradas do período de florescimento até a fase de produção de sementes. A inoculação com a estirpe Hup+ aumentou o total de N acumulado pelas plantas e a produção de sementes da soja cultivada sob luminosidade plena em comparação a estirpe Hup-. Após o florescimento, uma maior proporção de N-ureídos foi transportada na seiva xilemática das plantas inoculadas com a estirpe Hup+. O sombreamento das plantas, aumentou a proporção relativa de ureídos no exsudato xilemático e os índices de colheita de biomassa e N da soja. O efeito do sombreamento no transporte de N-ureído na seiva foi mais acentuado nas plantas inoculadas com a estirpe Hup- (29W) e refletiu em um aumento na produção de grãos dessas plantas em comparação ao tratamento não sombreado. Na última etapa do trabalho, foram conduzidos dois experimentos utilizando mutantes isogênicas Hup+ e Hup- de B. japonicum inoculadas em soja. As plantas foram cultivadas sob luminosidade total ou sombreadas (70% de sombreamento). A presença da hidrogenase induziu a sintese de ureídos nos nódulos e aumentou sua proporção no exsudato xilemático das plantas. Os nódulos formados com a mutante Hup- apresentaram uma baixa eficiência relativa da fixação de N2 quando cultivada sob luminosidade total, no sombreamento, ocorreu um aumento na ER da nitrogenase. Nessas plantas, observou-se uma correlação positiva entre a ER e a proporção relativa de N- ureídos na seiva xilemática. Os resultados obtidos sugerem a utilização da porcentagem de N-ureído no exsudato xilemático, durante o período reprodutivo das plantas, como parâmetro para a avaliação da eficiência do sistema simbiótico e da atividade dos nódulos de soja. | pt_BR |

| dc.description.abstractOther | The hydrogen production by nitrogenase has been identified as oneotthe factors that limits the efficiency on N2 fixation. Soybean plants show positive correlation of the relative efficiencies (RE) with the percentage of ureide-N in the xilem sap and harvest indices. The presence of an active system allows greater efficiency in biological nitrogen fixation (BNF) and can increase grain yield in soybean plants. Increase in grain field can only be attained with a comprehensive understanding of the plant physiology and identification of the major factors limiting the rates and the duration of the process related to BNF. In this work, the effect of hydrogenase on N2 fixation, N transport and distribution in soybean plants was studied. In the first xiii experiment, soybean plants were inoculated with Bradyrhizobium japonicum strain: CB 1809 (BR 33) or with Bradyrhizobium elkanii strain: 29W (BR 29). We compared the effect of the Hup+ and Hup- phenotype on N2 fixation and N distribution in soybean shoots. The contribution of BNF was estimed using 15N isotope dilution technique. About 95% of the plant N was derived from BNF. The Hup+ strain fixed more N2 than the Hup- strain. The nitrogenase activity (ARA) of the plants inoculated with the Hup- strain was reduced after flowering in comparison to the Hup+ strain that maintained high rates of ARA during the seed filling stage. The higher activity nodules formed by the Hup+ strain was reflected in the greater proportion of ureide-N in xylem sap. Plants inoculated with Hup+ strain showed higher grain yield and N harvest indices than plants inoculated with the Hup- strain. In the second experiment, soybean plants were inoculated again with CB 1809 or 29W. The plants were grown up to yield under shading (30% full day light) or at full sunlight. Under full sunlight, seed yield of plants inoculated with the Hup+ strain were higher than plants inoculated with Hup- strain. For both strains, the shading increased the proportion of ureide-N transported to the shoot and the soybean harvest indices. The effect of shading on the soybean physiology was, however, more remarkable in plants inoculated with the Hup- strain that the ones inoculated with Hup+ strain. In the third experiment, soybean plants were inoculated with either Bradyrhizobium japonicum mutant Hup- (PJ17) or its isogenic revertant Hup+ (PJ17-1). The plants were grown up to flowering stage under shading (30% full daylight) or at full sunlight. The revertant Hup+ induced the ureide synthesis and increased its proportion in the soybean xylem sap when compared to the Hup- mutant. A significant increase in the RE of nodules formed by Hup- mutant was observed when plants were partially shaded. This increase in RE reflected in a greater proportion of fixed N transported as ureide. The significant differences in xylem sap composition (percentage ureide-N) observed under full daylight between the plant inoculated with Hup+ and Hup- mutants were not observed in shaded plants. A positive correlation between RE and percentage in xylem sap was observed in plants inoculated with Hup- mutant. The results indicate that under conditions of reduced carbon availability the Hup- strains can increase their efficiency. These results can help to explain the conflicting effects observed with the presence of Hup in Bradyrhizobium strains. The relative proportion of the ureide-N in the sap during the reproductive stage, could be used as a reliable parameter in soybean breeding programs indicating the efficiency of the symbiotic system and nodule activity. | en |
