dc.description.abstractNo presente estudo, testamos os extratos em metanol e suas partições da Annona tomentosa, espécie nativa do Brasil e popularmente conhecida como “araticum de moita” ou “araticum rasteiro”. O chá de suas folhas é utilizado na medicina popular como anti-inflamatório. Por esta razão, as atividades antinociceptiva e anti-inflamatória da A. tomentosa, de São Luís-MA, foram avaliadas em camundongos. Os extratos em metanol das cascas do caule (ATCM) e das folhas (ATFM) foram preparados por maceração em metanol e suas partições em hexano (ATFM-H), diclorometano (ATFM-D), acetato de etila (ATFM-Ac) e butanol (ATFM-Bu) foram preparadas por partição líquido-líquido. O efeito antinociceptivo dos extratos e suas partições foram avaliados pelos modelos de contorções abdominais induzidas por ácido acético e de retirada da cauda, enquanto os efeitos anti-inflamatórios foram avaliados pelos modelos do edema de pata e da bolsa de ar subcutânea. A partir do modelo de retirada da cauda foi avaliado o possível mecanismo de ação da partição ATFM-H utilizando antagonistas e inibidores, tais como naloxona, L-NAME, glibenclamida, atropina, naltrindol e nor-binaltorfimina. A migração leucocitária e a produção de TNF-α e IL-1β foram avaliadas a partir do modelo da bolsa de ar subcutânea estéril. O extrato em metanol ATFM (100 mg/kg) foi eficaz em reduzir as contorções abdominais em aproximadamente 49% (27,8 ± 4,4 contorções abdominais) e dentre suas partições, ATFM-H e ATFM-D reduziram as contorções abdominais em 42,3% (31,3 ± 5,9 contorções abdominais) e 34% (35,8 ± 7,7 contorções abdominais), respectivamente, em relação ao grupo controle negativo. ATFM-H apresentou efeito antinociceptivo central com envolvimento de receptores opioides do tipo delta, no modelo de retirada da cauda, e sem prejuízo da atividade locomotora espontânea dos animais no modelo do campo aberto, em relação ao grupo controle negativo. A partição ATFM-D reduziu significativamente o edema de pata induzido tanto por carragenina (100 mg/kg) em 39,3%, histamina (50 e 100 mg/kg) e serotonina (10, 50 e 100 mg/kg). O ATFM-D inibiu a migração de leucócitos totais para a bolsa de ar estéril, nas doses de 50 e 100 mg/kg, em 75,8% (3,91 ± 1,2 x 106/mL) e 92,9% (1,15 ± 0,5 x 106/mL), respectivamente, e a produção de TNF-α nas doses de 50 e 100 mg/kg em aproximadamente 43% (740 ± 85,7 pg/mL) e 67% (433,8 ± 52,1 pg/mL), respectivamente, em relação ao grupo controle PBS. O ATFM-D também reduziu a produção de IL-1β nas doses de 50 e 100mg/kg em aproximadamente 25% (452,3 ± 52,5 pg/mL) e 35% (388,7 ± 42,3 pg/mL), respectivamente, em relação ao grupo controle PBS. Conforme demonstrado pelos métodos empregados, podemos concluir que o extrato em metanol de folhas da A. tomentosa apresentou efeito antinociceptivo e anti-inflamatório nos modelos de dor e inflamação aguda, sugerindo suas potencialidades para fins terapêuticos.por

dc.description.abstractOtherIn this study, we tested the methanolic extracts and their partitions of Annona tomentosa, native specie of Brazil and popularly known as “araticum rasteiro” or “araticum de moita”. The tea from its leaves has been used as anti-inflammatory by native community. For this reason, antinociceptive and anti-inflammatory activities of A. tomentosa, from São Luís-MA, were evaluated in mice. The methanolic extracts from stem bark (ATCM) and leaves (ATFM) were prepared by maceration with methanol and their partitions from hexane (ATFM-H), dichloromethane (ATFM-D), ethyl acetate (ATFM-Ac) and butanol (ATFM-Bu) were prepared by liquid-liquid partition. The antinociceptive effect of extracts and fractions were evaluated by abdominal writhing and tail-flick tests, while the anti-inflammatory effects were evaluated by paw oedema and air pouch tests. From the tail flick, we evaluated by the possible action mechanism of A. tomentosa using antagonists and inhibitors, such as naloxone, L-NAME, glibenclamide, atropine, naltrindole and nor-binaltorphimine. The leukocyte migration and production of TNF-α and IL-1β were evaluated from the air pouch test. The methanolic extract ATFM (100 mg/kg) reduced abdominal writhing by aproximately 49% (27,8 ± 4,4 abdominal writhing). Among its fractions only ATFM-H and ATFM-D were able to reduce the abdominal writhing 42,3% (31,3 ± 5,9 abdominal writhing) and 34% (35,8 ± 7,7 abdominal writhing), respectively, in relation to negative control group. The ATFM-H showed central antinociceptive effect from the involvement of the delta opioid receptors, in the tail flick test, without promote alterations in the locomotor activity of the mice in the open field test, in relation to negative control group. The partition ATFM-D was effective in decreasing paw oedema induced by carrageenan (100 mg/kg), histamine (50 and 100 mg/kg) and serotonine (10, 50 and 100 mg/kg). O ATFM-D inhibited leukocyte migration to air pouch, at doses 50 and 100 mg/kg, in 75,8% (3,91 ± 1,2 x 106/mL) and 92,9% (1,15 ± 0,5 x 106/mL), and TNF-α production at doses of 50 and 100 mg/kg by approximately 43% (740 ± 85,7 pg/mL) and 67% (433,8 ± 52,1 pg/mL), respectively, in relation to PBS control group.
