dc.description.abstractA Borreria verticillata (L.) G. Mey é uma planta herbácea da família Rubiaceae, considerada uma erva daninha e conhecida como vassourinha-de-botão. Por ser usada pela população como analgésica e antipirética, o objetivo deste trabalho foi avaliar o potencial antinociceptivo e anti-inflamatório do extrato aquoso das folhas de B. verticillata (EFBV), das frações flavonoídica (Flav), butanólica (BuOH) e acetato de etila (AcOEt) e dos compostos rutina (Ru, rendimento de 58%) e ácido clorogênico (AC, rendimento de 3%), administrados via oral em camundongos. Estudos anteriores, mostraram a atividade do extrato de EFBV, do flavonóide rutina e do composto fenólico ácido clorogênico em diversas atividades biológicas, incentivando nossa pesquisa com estes compostos, objetivando a confirmação dos usos da B. verticillata. A avaliação da atividade antinociceptiva pelo modelo das contorções abdominais (ácido acético), mostrou que a administração de EFBV (100, 300 e 1000mg/kg) produziu inibições no número de contorções de 23,6%, 30,3% e 47,3% respectivamente, quando comparados ao veículo, assim como o controle positivo (CP) indometacina reduziu as contorções. No modelo de imersão cauda, a administração de EFBV não mostrou alterações no tempo de latência dos animais diante do estímulo térmico e para confirmar estes resultados antinociceptivos, avaliamos a atividade motora do EFBV, no qual sua administração não modificou o tempo de permanência dos animais no rota-rod. A avaliação da atividade anti-inflamatória, utilizando o modelo de pleurisia por carragenina, mostrou que a administração de EFBV (1g/kg) inibiu a migração leucocitária em 33,7%, comparativamente ao veículo e as frações Flav (20mg/kg), BuOH (200mg/kg) e AcOEt (25mg/kg) também apresentaram reduções na migração leucocitária de 38,9%, 71,1%, 78,8% respectivamente. Avaliando a atividade antiedematogênica com o modelo de edema de orelha (óleo de cróton), a administração de EFBV (1g/kg) reduziu o edema de orelha em 47,6%. Na artrite articular por zimosan, o EFBV (1g/kg), a Ru (30mg/kg) e o AC (2mg/kg) reduziram o edema da articulação fêmur-tibial em 57,8%, 49,2% e 51,7% respectivamente, quando comparados aos veículos. Neste mesmo modelo, o EFBV (1g/kg), os compostos Ru (30mg/kg) e AC (2mg/kg) inibiram a migração leucocitária para cavidade articular em 64,9%, 79,5% e 77,8%, quando comparados aos seus respectivos veículos, assim como a CP dexametasona também produziu reduções nos dois parâmetros avaliados neste modelo. Na mensuração do diâmetro fêmur-tibial, seguida da dosagem da citocina TNF-α no macerado articular, EFBV, Ru e AC reduziram a espessura articular em 58,1% (EFBV, Ru) e 40,7% (AC), quando comparados ao controle salina-zimosam. Avaliamos a concentração de TNF-α após administração de EFBV, Ru ou AC, onde observamos reduções de 30,4%, 37,0%, 21,7%, respectivamente, na concentração da citocina, comparados com controle salina-zimosan e semelhante ao CP, reduzindo em 50,0% esta concentração. Estes resultados evidenciam que a antinocicepção do EFBV, no modelo das contorções abdominais mostrando envolvimento de mecanismos anti-inflamatórios, tendo este extrato apresentado atividade antiedematogênica no modelo de edema de orelha e, juntamente com os compostos Ru e AC, no modelo de artrite por zimosan. Adicionalmente, o EFBV e as frações (Flav, BuOH e AcOEt) inibiram a migração leucocitária na pleurisia por carragenina, tendo também o extrato, Ru e AC apresentado efetividade semelhante na artrite articular por zimosan, reduzindo também a concentração de TNF-α. Esses resultados sugerem que os efeitos anti-inflamatório, antinociceptivo e anti-artrite obtidos envolve a inibição do TNF-α, justificando os efeitos biológicos do EFBV, da rutina e do ácido clorogênico, podendo explicar alguns dos usos da B. verticillata (L.) G. Mey.por

dc.description.abstractOtherBorreria verticillata (L.) G. Mey is a herbaceous plant of the family Rubiaceae, considered a weed and known as vassourinha-de-botão. Because it is used in the population as analgesic and antipyretic, the objective of this work was to evaluate the antinociceptive and anti-inflammatory potential of B. verticillata (EFBV) aqueous extract of the leaves, flavonoid (Flav), butanol (BuOH) and ethyl acetate (AcOEt) fractions and compounds rutin (Ru, 58% yield) and chlorogenic acid (AC, 3% yield), administered orally in mice. Previous studies have shown the activity of the EFBV extract, the flavonoid rutin and the phenolic compound chlorogenic acid in several biological activities, encouraging our research with these compounds, aiming to confirm the uses of B. verticillata. The evaluation of antinociceptive activity by the writhing test (acetic acid), showed that the administration of EFBV (100, 300 and 1000mg/kg) produced inhibitions in the number of writhes of 23.6%, 30.3% and 47.3%, respectively, when compared to the vehicle, as well as the positive control (CP) indomethacin reduced of the writhes. In the tail-immersion test, the administration of EFBV did not show changes in the latency time of the animals in front of the thermal stimulus and to confirm these antinociceptive results, we evaluated the motor activity of the EFBV, in which its administration did not modify the dwell time of the animals in the rota-rod. The evaluation of anti-inflammatory activity through the carrageenan-induced pleurisy test showed that EFBV (1g/kg) inhibited leukocyte migration in 33.7%, compared to the vehicle and the fractions Flav (20mg/kg), BuOH (200mg/kg), AcOEt (25mg/kg) presented reductions in the migration of 38.9%, 71.1%, 78,8% and 85.5% respectively. Evaluating the antiedematogenic activity with the ear edema test (croton oil), the administration of EFBV (1g/kg) reduced edema by 47.6%. In zymosan-induced articular arthritis, EFBV(1g/kg), Ru (30mg/kg) and AC (2mg/kg) reduced edema of the femoral-tibial joint in 57.8%, 49.2% and 51.7%, respectively, when compared to vehicles. In the same test, EFBV (1g/kg), Ru (30mg/kg) and AC (2mg/kg) compounds inhibited the leukocyte migration to the joint cavity in 64.9%, 79.5% and 77.8%, when compared to their respective vehicles and CP dexamethasone also produced reductions in the two parameters evaluated in this model. In the measurement of the femoral-tibial diameter, followed by cytokine TNF-α dosing in the joint macerate, EFBV, Ru and AC reduced joint thickness by 58.1% (EFBV, Ru) and 40.7% (AC), when compared to saline-zymosan control, with CP dexamethasone reduced the thickness by 76.5%. The concentration of TNF-α after administration of EFBV, Ru or AC, where we observed reductions of 30.4%, 37.0%, 21.7%, respectively, in the cytokine concentration, compared to saline-zymosan control and similar to CP, reducing this concentration by 50.0%. These results show that the antinociception of the EFBV, in the writhing test, participate in anti-inflammatory mechanisms, and this extract showed antiedematogenic activity (ear edema) and, together with the compounds Ru and AC, at the femoral-tibial joint in arthritis by zimosan. In addition, EFBV and fractions (Flav, BuOH and AcOEt) inhibited leukocyte migration in pleurisy by carrageenan, and the extract, Ru and AC also showed similar effectiveness in zymosan-induced arthritis, also reducing the concentration of TNF- α. These results suggest that the anti-inflammatory, antinociceptive and anti-arthritis effects involved the inhibition of TNF-α, justifying the biological effects of EFBV, rutin and chlorogenic acid, and may explain some of the uses of B. verticillata (L.) G. Meyeng
