dc.description.abstractA criptosporidiose é uma infecção intestinal e respiratória causada por Protistas parasitos do gênero Cryptosporidium que infectam uma ampla variedade de hospedeiros em todas as classes de vertebrados. A importância das vias de transmissão na epidemiologia da criptosporidiose não é totalmente clara, entretanto, na última década várias técnicas de biologia molecular foram desenvolvidas para diagnosticar espécies de Cryptosporidium, genótipos e subtipos. Estas técnicas estão sendo utilizadas para estudos epidemiológicos em áreas endêmicas e epidêmicas, o que melhorou significativamente a compreensão da transmissão de criptosporidiose em seres humanos e animais. Neste contexto, o presente estudo teve como objetivos: diagnosticar por meio do exame parasitológico de fezes de cães e gatos domiciliados a presença de oocistos de Cryptosporidium spp.; caracterizar genotipicamente as espécies do gênero através do sequenciamento nucleotídico e realizar análises filogenéticas, comparando as sequências obtidas com aquelas sequências depositadas no GenBank. No total, 119 amostras fecais foram coletas de residências pertencentes ao bairro de Campo Grande, Zona Oeste do município de Rio de Janeiro, Brasil. Oocistos de Cryptosporidium foram detectados nas amostras de 5/64 (7,8%) cães e 3/55 (5,4%) gatos. Após a realização do sequenciamento, C. canis (n = 3) e C. parvum (n = 2) foram às espécies identificadas em cães e C. felis (n = 3) foi detectada em gatos. As três espécies diagnosticadas neste estudo são consideradas de importância em saúde humana e animal. Além da criptosporidiose ser debilitante para o hospedeiro, esses dados refletem preocupação, pois as espécies diagnosticadas, apresentam potencial zoonótico. Além da importância do diagnóstico de espécies de Cryptosporidium neste estudo, outros parasitos gastrointestinais foram detectados e não devem ser negligenciados, visto que também apresentam importância tanto em saúde humana quanto animal. Os resultados obtidos neste estudo constituíram primeiro registro de parasitismo por C. parvum em cães confirmado no Brasil e consequentemente, no estado do Rio de Janeiro através do uso de ferramentas moleculares, contribuindo para traçar a epidemiologia molecular deste agente etiológico mundialmentepor

dc.description.abstractOtherThe cryptosporidiosis is a intestinal and respiratory infection caused by parasites protists of the genus Cryptosporidium that infect a wide host range in all classes of vertebrates. The importance of transmission routes in the epidemiology of cryptosporidiosis is not totally clear, however, in the last decade several techniques of molecular biology have been developed to diagnose species of Cryptosporidium, genotypes and subtypes. These techniques are being used for epidemiological studies in endemic and epidemic areas, which has significantly improved the understanding of the transmission of cryptosporidiosis in humans and animals. In this context, the present study had the following objectives: to diagnose the presence of Cryptosporidium spp. oocysts by parasitological examination of faeces of dogs and cats domiciled; genotyping the species of the genus through nucleotide sequencing and performing phylogenetic analyzes, comparing the sequences obtained with those sequences deposited in GenBank. In total, 119 fecal samples were collected from residences belonging to the Campo Grande neighborhood, West Zone of the city of Rio de Janeiro, Brazil. Cryptosporidium oocysts were detected in the samples of 5/64 (7.8%) dogs and 3/55 (5.4%) cats. After the sequencing, C. canis (n = 3) and C. parvum (n = 2) were identified in dogs and C. felis (n = 3) was detected in cats. The three species diagnosed in this study are considered of importance in human and animal health. Besides cryptosporidiosis is debilitating for the host, these data reflect concern, because the species diagnosed have a zoonotic potential. In addition to the importance of the diagnosis of Cryptosporidium species in this study, another gastrointestinal parasites were detected and should not be neglected, since they are also important in both human and animal health. The results obtained in this study constituted the first registry of parasitism by C. parvum in dogs confirmed in Brazil and consequently in the state of Rio de Janeiro through the use of molecular tools, contributing to trace the molecular epidemiology of this etiological agent worldwideeng
