dc.description.abstractNeste trabalho, foram investigados os efeitos da azadiractina A no curso da infecção por Trypanosoma cruzi no intestino do triatomíneo vetor Rhodnius prolixus. No Rhodnius prolixus o desenvolvimento do clone Dm-28c do T. cruzi decresce de uma maneira dose dependente, e a ED50 deste efeito inibitório da azadiractina de 0.25 ug/ml de sangue. Usando este inseto, nós conduzimos experimentos de longo prazo demonstrando que a azadiractina (1.0 ug/ml de sangue) bloqueia o desenvolvimento do T. cruzi durante 120 dias após o tratamento com a droga. Similarmente, a eliminação do T. cruzi nas fezes e urina foi bloqueada por um período de 50 dias após a infecção dos insetos tratados com azadiractina. Larvas de 5º estádio de R. prolixus infectadas com T. cruzi realizam uma drástica inibição do desenvolvimento do tripanosomatídeo quando tratadas com azadiractina A. A discussão destes resultados enfoca a possibilidade de que a azadiractina deve atuar diretamente na fisiologia intestinal e/ou indiretamente através do sistema neurosecretório.por

dc.description.abstractOtherStudies were carried out the effects of azadirachtin on the course of Trypanosoma cruzi infection in the gut of triatomine vector, Rhodnius prolixus. In Rhodnius prolixus the development of T. cruzi clone Dm-28c decreased in a dose dependent manner, and ED50 of this inhibitory effect was 0.25 ug azadirachtin/ml bloodmeal. Using this insect, we conducted a long term experiment which showed that azadirachtin (1.0 ug/ml bloodmeal) completely blocks the development of T. cruzi even 120 days after treatment with the drug, and after four infectant meals. Similarly, the elimination of T. cruzi in feces and urine was also completely blocked over a period of 50 days after infection in insects treated with azadirachtin. Fifth-instar larvae of Rhodnius prolixus infected with T. cruzi displayed drastic inhibition of trypanosome development when treated with azadirachtin. The discussion of these results focuses on the possibility that azadirachtin may act directly on gut physiology and/or indirectly throught the neurosecretory system.por
