dc.description.abstractO presente trabalho teve como objetivo estudar a composição química do óleo essencial de Thymus vulgaris (tomilho) usando diferentes técnicas de extração. As técnicas de extração utilizadas foram: hidrodestilação (HD), extração com solvente (SE), extração com fluído supercrítico (SFE) e microextração em fase sólida (SPME) e a identificação dos componentes foi realizada utilizando GC-MS. Foram investigadas algumas variáveis que podem influenciar no processo de extração, tais como perfil de temperatura, tempo de extração e escolha da fibra para SPME, e pressão e tempo de extração para SFE. Os métodos de extração utilizados mostraram-se eficientes para identificação dos principais componentes presentes no tomilho, porém com algumas diferenças qualitativas e quantitativas. Quantitativamente, os componentes majoritários, em todas as técnicas utilizadas, foram monoterpenos e sesquiterpenos. Entre os métodos convencionais a extração com solvente apresentou maior quantidade de componentes extraídos (30) quando comparado com a hidrodestilação, que extraiu apenas 24 compostos. A extração com solvente também apresentou maior concentração de alguns componentes presentes nas folhas de Thyumus vulgaris quando comparado com a hidrodestilação. Comparando a extração usando CO2 supercrítico com a extração com solvente, a primeira mostrou-se mais eficiente, pois extraiu uma maior quantidade de compostos (33) em um menor período de tempo (60min). Além disso, os extratos obtidos através do CO2 em alta pressão são livres de solvente, o que aumenta a pureza/qualidade do óleo essencial. A SPME, além de ser o procedimento de extração mais simples foi o mais rápido (30min), e ainda mostrou-se capaz de identificar um maior número de compostos de T. vulgaris (47) quando comparado aos demais métodos utilizados. Ademais, a microextração em fase sólida indicou a presença dos terpenos β-terpineno, α-Cubebene e Cadalene, ainda não mencionados em outros trabalhos de pesquisa com o Thymus vulgaris.por

dc.description.abstractOtherThis work aim to study the chemical composition of leaves of Thymus vulgaris (thyme) using different extraction techniques. The extraction techniques used were: hydrodistillation (HD), solvent extraction (SE), supercritical fluid extraction (SFE) and solid phase microextraction (SPME). The identification of the components was performed using GC-MS. Some variables that can influence the extraction process were investigated, such as temperature profile, extraction time and choice of fiber for SPME and pressure and extraction time for SFE. The extraction methods used were effective for identifying key components present in thyme, but with some qualitative and quantitative differences. Quantitatively, the major components in all techniques were monoterpenes and sesquiterpenes. Among the conventional methods, extraction solvent obtained a higher amount of extracted components (30) when compared with hydrodistillation, with only 24 compounds. The solvent extraction showed higher concentrations of some components in Thyumus vulgaris leaves when compared with hydrodistillation. Comparing the supercritical fluid extraction and solvent extraction, the first was more efficient because obtained a greater range of extracted compounds (33) in a shorter period of time (60 min). Furthermore, the extracts obtained by SFE are free of solvent, which increases the purity/quality of the oil. The SPME is an extraction procedure easier and faster (30min), besides being even able to identify a greater number of compounds of T. vulgaris (47), when compared to other methods. Moreover, the solid phase microextraction indicated the presence of compounds β-terpinene, α- Cubebene and Cadalene. These three compounds were not mentioned in others researches with Thymus vulgaris.eng
