dc.description.abstract | O abuso de esteróides anabólicos androgênicos (EAA), dentro e fora do cenário esportivo, se constitui atualmente em grande problema de saúde pública. Embora, estudos relativos a alterações comportamentais e hidroeletrolíticas induzidas por EAA sejam bem conhecidos, o mesmo já não se observa quando esses parâmetros são analisados em conjunto (no mesmo estudo). Além disso, alterações tardias (na fase adulta) induzidas pelo uso abusivo de EAA na adolescência são ainda mais raras. Assim, o principal objetivo deste estudo, foi avaliar possíveis alterações comportamentais, hidroeletrolíticas e autonômicas de ratos adultos sedentários ou fisicamente ativos submetidos ao tratamento com EAA na fase juvenil. O presente estudo foi submetido ao Comitê de Ética da Universidade Federal Rural do Rio de Janeiro (CEPBE/IB/UFRRJ-014/2008), e seguiu as normas propostas pelo “Guide for the Care and Use of Laboratory Animals” (Instituto Nacional de Saúde Norte Americano). Foram utilizados 40 ratos Wistar jovens (40 dias) e 10 casais de residentes (90 dias). Os animais foram divididos aleatoriamente nos seguintes grupos: tratados com proprionato de testosterona (Grupo PT) na dose de 5mg/kg/dia (i.m.) ou veículo (óleo de milho) no mesmo volume e via de administração (Grupo Controle, CTR) por 5 semanas ininterruptas. Ambos os grupos foram subdivididos em sedentários (PTsed ou CTRsed) e fisicamente ativos (PTexe ou CTRexe). Na 4a semana de administração (9 semanas de idade), os animais foram submetidos ao teste de agressividade e os resultados mostraram aumento significativo deste parâmetro (P < 0,05) no grupo PTsed em relação ao seu controle (CTRsed). Curiosamente, o aumento da agressividade foi amplamente inibido pelo exercício físico resistido, uma vez que não foram observadas diferenças significativas nos parâmetros que avaliam agressividade (ataques efetivos e ameaças) entre os grupos PTexe e CTRexe (P < 0,05) sendo ambos similares aos do grupo controle CTRsed. No teste do campo aberto houve aumento significativo tanto da exploração horizontal (número de quadrados percorridos/5 min.) como na vertical (número de rearings/5 min.) e diminuição no tempo de imobilidade no grupo PTsed comparado ao CTRsed (P < 0,05). Mais uma vez, o EFR foi capaz de reverter o efeito produzido pelo EAA, pois não houve diferença nestes parâmetros entre os grupos PTexe e CTRexe (P < 0,05) sendo ambos similares aos do grupo controle CTRsed. No teste do labirinto em cruz elevado não houve alteração significativa no tempo de permanência em nenhum dos braços entre os grupos, mas, as freqüências de entradas nos braços tiveram significativas diferenças por o grupo tratado PTsed ter maior número significativo que do seu respectivo controle CTRsed em ambos os braços, o que sugere inibição do efeito ansiolítico nos protocolos de administração de EAA e de EFR utilizados neste estudo. No estudo hidroeletrolítico houve aumento significativo (P < 0,05) na ingestão cumulativa de água e sódio no CTRexe, a partir de 60 minutos e assim por diante até o término do teste, em relação aos demais grupos em condições basais. O grupo tratado com EAA e que praticou exercício físico (PTexe) não apresentou este efeito, o que sugere que o PT foi capaz de prevenir as respostas adaptativas ao EFR. No estudo da variabilidade da frequência cardíaca observou-se que o tratamento com EAA aumentou a modulação simpática (diminuiu a parassimpática) no grupo sedentário tratado com PT e que este efeito, foi totalmente bloquedo pelo EFR, pois não houve diferença no balanço simpato-vagal cardíaco entre os grupos PTexe e CTRexe (P < 0,05) sendo ambos similares aos do grupo controle CTRsed. O tratamento com EAA na fase juvenil (adolescência) aumentou a agressividade e a atividade exploratória dos ratos na fase adulta e o protocolo de exercício utilizado foi efetivo em prevenir tais alterações. Além disso, as respostas hidroeletrolíticas adaptativas de aumento de ingestão de fluidos induzidos pelo exercício foram bloqueadas pelo uso de EAA na adolescência. É possível que este efeito mal adaptativo observado nos animais fisicamente ativos tratados com EAA, possa ser explicado, pelo menos em parte, pelo aumento da modulação simpática, conhecidamente retentora de sal e água observada na análise espectral. dc.description.abstractOther | The abuse of anabolic androgenic steroids (AAS), and off the sports scene, it is currently in big public health problem. Although studies on behavioral changes induced by AAS and electrolyte are well known, the same is not observed when these parameters are analyzed together (in the same study). In addition, late changes (in adulthood) induced by the use of AAS abuse in adolescence are even more rare. Thus, the main objective of this study was to assess possible behavioral changes, electrolyte and rat autonomic sedentary or physically active adults undergoing treatment with AAS in the juvenile phase. This study was submitted to the Ethics Committee of University Rural of Rio de Janeiro (CEPBE/IB/UFRRJ-014/2008), and followed the guidelines proposed by the "Guide for the Care and Use of Laboratory Animals" (National Institute of North American Health). A total of 40 young Wistar rats (40 days) and 10 pairs of residents (90 days). The animals were randomly divided into the following groups: proprionate treated with testosterone (PT) at a dose of 5mg/kg/day (im) or vehicle (corn oil) in the same volume and route of administration (control group, CTR) for 5 weeks uninterrupted. Both groups were subdivided into sedentary (or PTsed CTRsed) and physically active (or PTexe CTRexe). In the fourth week of administration (9 weeks of age), animals were tested for aggressiveness and the results showed a significant increase of this parameter (P <0.05) in PTsed in relation to its control (CTRsed). Interestingly, the increased aggressiveness was largely inhibited by resistive exercise, since there were no significant differences in parameters that assess aggression (actual attacks and threats) between the groups and PTexe CTRexe (P <0.05) and both are similar to CTRsed the control group. In the open field test showed a significant increase both the horizontal scanning (number of squares crossed / 5 min.) And vertical (number of rearings / 5 min.) And decrease in the immobility time compared to CTRsed PTsed group (P <0 , 05). Again, the EFR was able to reverse the effect produced by the AAS, as there was no difference in these parameters between groups and PTexe CTRexe (P <0.05) and both are similar to the control group CTRsed. In testing the elevated plus maze, no significant change in parameter the arms in any group and which probably indicates absence of anxiolytic in management protocols and EFR AAS used in this study. Electrolyte in the study increased significantly (p <0.05) in cumulative intake of water and sodium CTRexe from 60 minutes and so on until the end of the test, compared to other groups at baseline. The group treated with AAS and practiced physical exercise (PTexe) did not show this effect, suggesting that the PT was able to prevent the adaptive responses to the EFR. In the study of heart rate variability was observed that AAS treatment increased sympathetic modulation (decreased parasympathetic) in the sedentary group treated with PT and that this effect was totally blocked by EFR because there was no difference in cardiac vagal-sympathetic balance between groups and PTexe CTRexe (P <0.05) and both are similar to the control group CTRsed. The AAS treatment in the juvenile phase (adolescence) increased aggressiveness and exploratory activity of rats in adulthood and the exercise protocol used was effective in preventing such changes. Furthermore, the adaptive responses electrolyte of increased fluid intake induced by exercise have been blocked by the use of EAA in adolescence. It is possible that this effect maladaptive observed in animals treated with AAS physically active, may be explained at least in part, by increasing sympathetic modulation, known to salt and water retaining observed in the spectral analysis.
