The objective of this study was to investigate the effect of training interruptions of the annual training cycle of eventing horses on the main conditioning parameters V200 and VLA4; and on the enzymatic serum profile of AST, CK and LDH, as well as on uric acid, glucose, lactate, and heart rate. Eighteen Brazilian Sport horses at the beginning of the training season for eventing were selected. The animals were divided throughout the training season in groups 1, without training interruptions, and group 2, with training interruptions. A standardized effort test was done at the beginning and at the end of the season, with a 25-minute duration and reaching top speeds of 9m/s. The heart rate was analyzed through a Polar Equine heart rate monitor, while the blood parameters were obtained through a catheter on the left jugular vein. The V200 conditioning parameter was calculated using linear regression, while VLA4 was calculated through exponential regression. All training sessions were overseen, with any interruptions have its cause and duration registered. The statistical analyzes were done through the R program, in version 4.1.2. All horses in group 2 had interruptions longer than 10 days in their training programs, with the most common cause being change in equestrian discipline, followedby several veterinary causes. There was a significant difference between both groupsregardingdays without training (p<0,0005), with mean values of 45 and 87 days for horses in Groups 1 and 2, respectively. The values recorded for V200 in the first test were 10,02 ± 1,04m/s and 9,58± 0,89m/s for the first and second groups, respectively, while for the second test the values were 10,24 ± 1,01m/s and 9,52 ± 0,77m/s. For VLA4, in the first test, the values were of 6,75 ± 2,09m/s and 6,41 ± 2,67m/s for groups 1 and 2, respectively, while for the second test the values were of 7,51 ± 2,08m/s and 5,86 ± 1,57m/s. When comparing both conditioning parameters V200 and VLA4, no difference (p>0,05) was observed between both groups and both tests. Meanwhile, the uric acid indicated a better physical conditioning fitness in the horses without interruptions, also showing an improvement in both group throughout the year. The muscle enzymes AST, CK, and LDH stayed in the reference range, indicting the lack of muscle damage during the training sessions and effort tests. The glucose determination showed a period of marked hyperglycemia after the exercise, with fluctuations throughout due to feeding and supplements. In conclusions, there was no significant difference between the main physical conditioning parameters regarding the groups with or without training interruptions, indicating that the break period of 10 days was not enough to cause a change in the horse’s fitnesspt_BR
