dc.description.abstractPentatrichomonas hominis ou Trichomonas hominis como era chamado, é uma espécie de protozoário flagelado, anaeróbio, unicelular, amitocondrial pertencente aos Trichomonadidae, reconhecida morfologicamente pela presença de cinco flagelos anteriores e um único flagelo recorrente, inicialmente considerado comensal. Habita principalmente o intestino grosso de vários animais, incluindo gatos, cães e primatas não humanos. No entanto, alguns estudos têm documentado uma associação entre diarreia e a presença de P. hominis em cães e gatos, doença gastrointestinal e até associado a doenças respiratórias em humanos, o que aumenta a possibilidade de que P. hominis possa representar uma fonte de infecção com potencial zoonótico, ao invés de ser um organismo comensal. O isolamento de P. hominis em novos hospedeiros, como coruja, cabras, búfalos e porcos indica que este flagelado pode ter se adaptado a novos hospedeiros. No Brasil, poucos estudos foram conduzidos com o propósito de identificar as espécies que podem parasitar animais domésticos e suas interações com o hospedeiro. O objetivo do presente estudo foi relatar, pela primeira vez no Brasil, a presença do protozoário flagelado Pentatrichomonas hominis parasitando naturalmente o trato reprodutivo da espécie bovina. Adicionalmente, a eficiência de duas técnicas de coleta de muco vaginal para diagnóstico de tricomonose bovina foi comparada. Para tanto, a tese foi dividida em dois capítulos, sendo que o primeiro teve como objetivo principal relatar a presença do P. hominis em lavado prepucial de três touros da raça Nelore. Assim, para o diagnóstico, amostras de muco vaginal e lavado prepucial foram coletadas em meio Lactopep e posteriormente cultivadas in vitro em meio Hanks, e repicadas a cada 72 horas. As amostras do cultivo que apresentaram presença de organismos móveis foram submetidas a análises moleculares pela técnica da reação em cadeia de polimerase e, para confirmação final do diagnóstico, foi utilizada a técnica do polimorfismo no comprimento do fragmento de restrição e sequenciamento. O segundo capítulo comparou duas técnicas de coleta de muco vaginal, uma comumente utilizada e recomendada pela maioria dos guias atuais de coleta de material biológico e outra utilizando a escova citológica, amplamente utilizada na medicina humana e ainda, em citologias da espécie equina. Trinta fêmeas da raça Nelore em idade reprodutiva foram selecionadas de forma aleatória e submetidas às duas técnicas de coleta de muco vaginal: swab estéril descartável (Técnica 1) e a escova ginecológica estéril descartável (Técnica 2). Após avaliações qualitativas e do cultivo celular, foi possível concluir que seis amostras coletadas apresentavam estruturas móveis no cultivo, logo o uso da escova citológica trouxe o benefício de conseguir coletar quantidade maior de material biológico, o que pode facilitar e aumentar as chances de diagnóstico de parabasalídeos.pt_BR

dc.description.abstractOtherPentatrichomonas hominis or Trichomonas hominis as it was called, is a species of flagellated, anaerobic, unicellular, amitochondrial protozoan belonging to the Trichomonadidae, morphologically recognized by the presence of five anterior flagella and a single recurrent flagellum, initially considered commensal. It mainly inhabits the large intestine of various animals, including cats, dogs, and non-human primates. However, some studies have documented an association between diarrhea and the presence of P. hominis in dogs and cats, gastrointestinal disease and even associated with respiratory diseases in humans, which increases the possibility that P. hominis may represent a source of infection. with zoonotic potential, rather than being a commensal organism. The isolation of P. hominis in new hosts, such as owls, goats, buffaloes and pigs indicates that this flagellate may have adapted to new hosts. In Brazil, few studies have been conducted with the purpose of identifying the species that can parasitize domestic animals and their interactions with the host. The aim of the present study was to report, for the first time in Brazil, the presence of the flagellated protozoan Pentatrichomonas hominis naturally parasitizing the reproductive tract of bovine species. Additionally, the efficiency of two vaginal mucus collection techniques for the diagnosis of bovine trichomonosis was compared. Therefore, the thesis was divided into two chapters, the first of which aimed to report the presence of P. hominis in preputial lavage of three Nelore bulls. Thus, for diagnosis, samples of vaginal mucus and preputial lavage were collected in Lactopep medium and later cultured in vitro in Hanks medium, and subcultured every 72 hours. The culture samples that showed the presence of mobile organisms were subjected to molecular analysis by the polymerase chain reaction technique and, for final confirmation of the diagnosis, the technique of polymorphism in the length of the restriction fragment and sequencing was used. The second chapter compared two techniques for collecting vaginal mucus, one commonly used and recommended by most current guides for collecting biological material and the other using the cytological brush, widely used in human medicine and also in equine cytology. Thirty Nellore females of reproductive age were randomly selected and submitted to two techniques of vaginal mucus collection: sterile disposable swab (Technique 1) and sterile disposable gynecological brush (Technique 2). After qualitative and cell culture evaluations, it was possible to conclude that six samples collected had mobile structures in the culture, so the use of the cytological brush brought the benefit of being able to collect a greater amount of biological material, which can facilitate and increase the chances of diagnosis of Parabasalids.pt_BR
