dc.description.abstractO presente estudo foi dividido em dois capítulos, sendo que o primeiro teve como objetivo correlacionar diferentes estratégias de suplementação de progesterona (P4) em fêmeas da raça Nelore com diferentes temperamentos e a concentração de cortisol. Adicionalmente, foi avaliado o efeito destas estratégias e do temperamento animal sobre a taxa de concepção. O segundo capítulo teve por objetivo avaliar a eficiência de diferentes estratégias para elevar a concentração da P4 circulante em fêmeas da raça Nelore sobre a funcionalidade do corpo lúteo (CL), taxa de concepção e sobre a perda gestacional. Para os dois experimentos, 939 vacas foram submetidas à sincronização da ovulação e inseminadas em tempo fixo (IATF). Após a IATF, os animais foram distribuídos em três grupos: P4LA (n=305) 150mg de progestágeno injetável de longa ação, 7 dias após a IATF; GnRH (n=306), 10μg de acetato de buserelina, 7 dias após IATF; Grupo Controle (n=328) sem aplicação de hormônio. Avaliações ultrassonográficas em modo B (folículo pré-ovulatório - FPO) foram realizadas no dia da IA e as avaliações Doppler colorido (CL) foram realizadas no dia da inseminação, 7 e 16 dias após. Amostras de sangue foram coletadas no dia da IATF assim como no 7 o e 16o dias após, para dosagem de cortisol e P4, respectivamente. No primeiro capítulo, no momento da inseminação, os animais foram pontuados em escore de acordo com o comportamento apresentado, por meio desta avaliação eles foram classificados em animais de temperamento excitável (EXC) ou adequado (ADQ). Animais classificados como excitáveis tiveram maiores concentrações de cortisol e apresentaram menores volumes de FPO e CL sete dias após a IATF. Não foi observada diferença entre as variáveis intensidade e número de pixels do CL além das concentrações de P4, sete dias após a inseminação. Entre os animais EXC, os grupos GnRH e P4LA apresentaram maiores concentrações de P4. Houve uma tendência de menor concentração de P4 nos animais EXC quando comparados com ADQ no grupo controle (p=0,06). Os animais do grupo controle classificados como EXC apresentaram menor perfusão vascular comparado ao ADQ (p=0,04). Entre os animais ADQ, os grupos GnRH e P4LA apresentaram menor perfusão vascular comparado ao controle (p=0,04). Entre os animais EXC, foi observada maior taxa de concepção para os grupos GnRH e P4LA comparado ao controle (p=0,01). No grupo controle, a taxa de concepção dos animais ADQ foi maior comparado com animais EXC (p=0,05). No experimento 2, não foi observada diferença significativa entre os tratamentos nas variáveis volume, intensidade e número de pixels do CL, assim como em relação a concentração de P4, 7 dias após a ovulação. A perfusão vascular do CL 16 dias após a ovulação foi menor nos grupos P4LA e GnRH (P<0,01). As concentrações plasmáticas de P4, 16 dias após a ovulação, foram maiores nos grupos P4LA e GnRH em relação ao grupo controle (P=0,04). Foi observada diferença na taxa de concepção (P=0,003) e uma tendência na perda gestacional (P=0,07) em função dos tratamentos. Conclui-se que que utilização de GnRH ou P4 sete dias após a ovulação afeta a vascularização do CL aumenta as concentrações plasmáticas de P4 dezesseis dias após a ovulação e melhora as taxas de concepção em animais excitáveis, sendo uma alternativa viável a fim de minimizar o impacto negativo do estresse e melhorar os índices reprodutivos.pt_BR

dc.description.abstractOtherThe present study was divided into two chapters, the first of which aimed to correlate different progesterone (P4) supplementation strategies in Nellore females with temperament and cortisol concentration. Additionally, the effect of these strategies and animal temperament on conception rate was evaluated. The second chapter aimed to evaluate the efficiency of different strategies to increase the concentration of circulating P4 in Nelores females on the functionality of the corpus luteum (CL), conception rate and on pregnancy loss. For the two experiments, 939 cows were submitted to ovulation synchronization and inseminated at a fixed time (TAI). After the TAI, the animals were divided into three groups: P4LA (n=305) 150mg of long-acting injectable progestagen, 7 days after the TAI; GnRH (n=306), 10μg buserelin acetate, 7 days after FTAI; Control Group (n=328) without hormone application. B-mode ultrasound assessments (preovulatory follicle - PFO) were performed on the day of AI and color Doppler (CL) assessments were performed on the day of insemination, 7 and 16 days later. Blood samples were collected on the day of the TAI as well as on the 7th and 16th days after, for cortisol and P4 measurements, respectively. In the first chapter, at the time of insemination, the animals were scored according to the behavior presented, through this evaluation they were classified as excitable (EXC) or suitable (ADQ) animals. Animals classified as excitable had higher concentrations of cortisol and lower volumes of OPF and CL, seven days after TAI. No significant difference was observed between the variables intensity and number of CL pixels in addition to P4 concentrations, seven days after insemination. Among EXC animals, the GnRH and P4LA groups showed higher concentrations of P4. There was a trend towards lower P4 concentration in EXC animals when compared with ADQ in the control group (p=0.06). Control group animals classified as EXC had lower vascular perfusion compared to ADQ (p=0.04). Among ADQ animals, the GnRH and P4LA groups showed lower vascular perfusion compared to the control (p=0.04). Among the EXC animals, a higher conception rate was observed for the GnRH and P4LA groups compared to the control (p=0.01). In the control group, the conception rate of ADQ animals was higher compared to EXC animals (p=0.05). In experiment 2, no significant difference was observed between treatments in the variables volume, intensity and number of CL pixels, as well as in relation to P4 concentration, 7 days after ovulation. CL vascular perfusion 16 days after ovulation was lower in the P4LA and GnRH groups (P<0.01). Plasma concentrations of P4, 16 days after ovulation, were higher in the P4LA and GnRH groups compared to the control group (P=0.04). There was a significant difference in the conception rate (P=0.003) and a trend in pregnancy loss (P=0.07) as a function of the treatments. It is concluded that the use of GnRH or P4 seven days after ovulation affects the vascularization of the CL, increases plasma concentrations of P4 sixteen days after ovulation and improves conception rates in excitable animals, being a viable alternative in order to minimize the negative impact of stress and improve reproductive indices.en
