dc.description.abstractOs cães deixaram de ser animais de companhia para se tornarem membros do núcleo familiar onde a preocupação com a sanidade e bem-estar destes indivíduos cada vez mais aumenta e atrelado a isso, a farmacologia também é impulsionada na busca de medicamentos cada vez mais eficazes e seguros para controlar infestações. Rhipicephalus sanguineus ou carrapato marrom dos cães, é um artrópode que possui alta relevância em saúde pública devido a sua alta capacidade de transmissão de doenças, bem como seu potencial irritativo e espoliativo seja na fase de larva, ninfa ou adulta. Atualmente, na maioria dos casos, o seu controle é feito com acaricidas sintéticos como o fipronil na sua grande maioria, porém os produtos naturais têm ganhado cada vez mais espaço não só pela sua eficácia, mas também, pela segurança como é o caso do monoterpeno carvacrol e do fenilpropanóide eugenol. O presente trabalho apresenta duas etapas onde a primeira é a adaptação e padronização de uma técnica de bioensaio para testar compostos acaricidas frente a fase de adulto não alimentado de R. sanguineus, que consistiu primeiramente na adaptação e comparação de três técnicas conhecidas para teste contra a fase larval que são Teste de Imersão Larval (TIL), a metodologia de Monteiro et al. (2012) com o pacote de larvas impregnado (PLI) e o Teste do Pacote com Larvas (TPL) que se provou superior quando comparada as outras, apresentando menor valor de CL50 de fipronil, que foi de 1236 μg/mL, contra 5598,8 μg/mL para TIL e 4834,8 μg/mL para PLI, sendo a técnica escolhida para a obtenção das CL50 de fipronil, eugenol e carvacrol utilizadas para a avaliação sinérgica contra todas as fases não alimentadas. Carvacrol e eugenol apresentaram excelente sinergismo farmacológico para todas as fases, inclusive a de adultos, fase esta que apresenta maior resistência quando comparada as fases imaturas. Os valores de coeficiente de sinergismo, não excederam 0,548, comprovando o potencial dessa interação. Em contrapartida, o fipronil apresentou incompatibilidade com eugenol e carvacrol, apresentando antagonismo e diminuição da mortalidade em todas as proporções de concentração utilizadas para ambas as moléculas. Estudos com a fase adulta de R. sanguineus são escassos, este trabalho foi o primeiro a desenvolver uma metodologia de teste com carrapatos adultos não alimentados e avaliar o sinergismo entre moléculas naturais e sintéticas, abrindo portas para futuros experimentos com o intuito de inserir cada vez mais componentes botânicos para o combate de ectoparasitos.pt_BR

dc.description.abstractOtherDogs are no longer companion animals to become members of the family where the concern for the health and well-being of these individuals increases and linked to this, pharmacology is also driven in the search for increasingly effective medicines and insurance to control infestations. Rhipicephalus sanguineus or brown dog tick, is an arthropod that has high public health relevance due to its high capacity for disease transmission, as well as its irritating and despoiling potential, whether in the larval, nymph or adult phase. Currently, in most cases, its control is done with synthetic acaricides such as fipronil in its vast majority, but natural products have gained more and more space not only for their effectiveness, but also for safety as is the case with monoterpene. carvacrol and the phenylpropanoid eugenol. The present work presents two steps where the first is the adaptation and standardization of a bioassay technique to test acaricide compounds against the non-fed adult stage of R. sanguineus, which consisted primarily in the adaptation and comparison of three known techniques for testing against the larval stage which are Larval Immersion Test (TIL), the methodology of Monteiro et al. (2012) with the impregnated package of larvae (PLI) and the Test of Package with Larvae (TPL) which proved to be superior when compared to the others, presenting a lower value of fipronil LC50, which was 1236 μg/mL, against 5598, 8 μg/mL for TIL and 4834.8 μg/mL for PLI, being the technique chosen to obtain the LC50 of fipronil, eugenol and carvacrol used for the synergistic evaluation against all unfed phases. Carvacrol and eugenol showed excellent pharmacological synergism for all stages, including adults, which stage has greater resistance when compared to immature stages. The synergism coefficient values did not exceed 0.548, proving the potential of this interaction. On the other hand, fipronil showed incompatibility with eugenol and carvacrol, showing antagonism and decreased mortality in all concentration ratios used for both molecules. Studies with the adult stage of R. sanguineus are scarce, this work was the first to develop a test methodology with unfed adult ticks and to evaluate the synergism between natural and synthetic molecules, opening doors for future experiments in order to insert each time more botanical components to combat ectoparasites.en
