| dc.description.abstract | O tomateiro (Solanum lycopersicum L.) é uma das hortaliças mais produzidas e consumidas no Brasil e no mundo e com grande importância social e econômica. É suscetível a várias doenças que podem acarretar grandes perdas em produtividade e qualidade dos frutos. Dentre estas, está a murcha de fusarium, causada pelo fungo Fusarium oxysporum f. sp. lycopersici (Scc.) Snyder e Hansen (Fol) que é uma das doenças mais limitantes à produção de tomateiro. Este estudo teve como objetivos: a) caracterizar, funcionalmente, estirpes de Bacillus spp.isoladas de tecidos de plantas de tomateiro, mantidas na coleção do Laboratório de Genética e Bíoquimica da Embrapa Agrobiologia, com potencial de controle antagônico contra Fol e identificar os possíveis mecanismos envolvidos no antagonismo; b) determinar, em testes in vitro, estirpes bacterianas com atividade antagônica significativa a Fol; c) avaliar o potencial das estirpes em promover o controle da murcha de fusário do tomateiro industrial em condições de casa-de-vegetação; e e) avaliar os efeitos benéficos de Bacillus spp. em plantas de duas cultivares de tomate industrial. Realizaram-se testes de antagonismo, in vitro, e selecionaram-se oito estirpes, que promoveram redução de pelo menos 50% o crescimento micelial de Fol: DEST 30, DEST 31, DEST 38, DEST 60, DEST 78, DEST 83, DEST 132 e DEST 138. Estas foram submetidas a testes para caracterização funcional e quanto à capacidade de produzir proteases, celulases, pectinases, sideróforos, HCN e quitinases. A partir dos resultados, selecionaram-se quatro estirpes, DEST 31, DEST 60, DEST 83 e DEST 132, que foram avaliadas em casa de vegetação visando o controle da murcha de fusarium em duas cultivares de tomate industrial, Heinz 9553 e Clause HM 7885, inoculadas com dois isolados de Fol FUS 2903 e FUS 04-1. Todas as oito estirpes pré-selecionadas demonstraram atividade proteolítica e produção de sideróforos; a maioria apresentou atividade celulolítica e pectinolítica, com exceção de DEST 30; nenhuma das estirpes produziu HCN ou quitinase nos métodos empregados. De forma geral, as estirpes testadas não foram eficientes em controlar a murcha de fusarium do tomateiro. No entanto, plantas de Heinz 9553 tratadas com estirpe DEST 132 ou com as estirpes DEST 02, DEST 31 e DEST 132 apresentaram respostas positivas e significativas quanto ao crescimento quando inoculadas com FUS 2903 e FUS 04.1, respectivamente. Observou-se também maior acúmulo de massa fresca em plantas de Clause HM 7885 inoculadas com DEST 60, DEST 83 e DEST 132. Plantas tratadas com DEST 132 e inoculadas com FUS 04.1 tiveram acréscimo no crescimento e no aporte de massa fresca. Apesar de, em geral, não ter sido observado efeitos significativos das estirpes de Bacillus spp. testadas sobre o controle da doença, houve efeitos discretos quanto ao acúmulo de massa e melhoria no crescimento de plantas inoculadas com o patógeno. | pt_BR |

| dc.description.abstractOther | Tomato (Solanum lycopersicum L.) is one of the most produced and consumed vegetables in Brazil and worldwide, with significant social and economic importance. It is susceptible to various diseases that can cause significant losses in productivity and fruit quality. Among these, Fusarium wilt, caused by the fungus Fusarium oxysporum f. sp. lycopersici (Scc.) Snyder and Hansen (Fol), is one of the most limiting diseases for tomato production. This study aimed to: a) functionally characterize strains of Bacillus spp. isolated from tomato plant tissues, maintained in the collection of the Laboratory of Genetics and Biochemistry at Embrapa Agrobiology, with potential for antagonistic control against Fol and identify the possible mechanisms involved in antagonism; b) determine, in vitro tests, bacterial strains with significant antagonistic activity against Fol; c) evaluate the potential of the strains to promote control of Fusarium wilt in industrial tomato plants under greenhouse conditions; and d) assess the beneficial effects of Bacillus spp. on plants of two industrial tomato cultivars. Antagonism tests were conducted in vitro, and eight strains were selected, which promoted at least a 50% reduction in the mycelial growth of Fol: DEST 30, DEST 31, DEST 38, DEST 60, DEST 78, DEST 83, DEST 132, and DEST 138. These strains were subjected to tests for functional characterization and their ability to produce proteases, cellulases, pectinases, siderophores, HCN, and chitinases. Based on the results, four strains were selected, DEST 31, DEST 60, DEST 83, and DEST 132, which were evaluated in a greenhouse to control Fusarium wilt in two industrial tomato cultivars, Heinz 9553 and Clause HM 7885, inoculated with two isolates of Fol FUS 2903 and FUS 04-1. All eight pre-selected strains demonstrated proteolytic activity and siderophore production; most showed cellulolytic and pectinolytic activity, except for DEST 30; none of the strains produced HCN or chitinase in the methods employed. Overall, the tested strains were not efficient in controlling Fusarium wilt of tomato plants. However, Heinz 9553 plants treated with DEST 132 or with strains DEST 02, DEST 31, and DEST 132 showed positive and significant growth responses when inoculated with FUS 2903 and FUS 04.1, respectively. There was also a greater accumulation of fresh mass in Clause HM 7885 plants inoculated with DEST 60, DEST 83, and DEST 132. Plants treated with DEST 132 and inoculated with FUS 04.1 had increased growth and fresh mass input. Although, in general, significant effects of the tested Bacillus spp. strains on disease control were not observed, there were discreet effects regarding mass accumulation and growth improvement of plants inoculated with the pathogen. | en |
