| dc.description.abstract | O presente trabalho teve por objetivo realizar a detecção e a caracterização molecular de bactérias da família Anaplasmataceae e Mycoplasmataceae em populações de pequenos mamíferos em áreas de Floresta Amazônica, no estado do Acre, Brasil. A importância desta investigação está pautada em bactérias intracelulares obrigatórias que participam nos ciclos silvestres como agentes zoonóticos, e, portanto, de interesse para a saúde humana, além de parasitos de importância para os animais domésticos, e a própria ecologia e ciclo de vida do animal hospedeiro. Asáreas de estudos se concentram em áreas de mata da floresta amazônica, na cidade de Rio Branco, estado do Acre, Brasil. Para o diagnóstico direto foram utilizadas 186 amostras de lâminas de esfregaço sanguíneo, oriundas de roedores, marsupiais e morcegos capturados previamente, visando a detecção e caracterização de bactérias da família Anaplasmataceae e Mycoplasmataceae. Amostras de parênquima hepático armazenadas em RNA Later® tiveram seu DNA extraído pelo kit “DNeasy Blood & Tissue” (Qiagen®) e foram utilizadas no diagnóstico molecular. Dentre as amostras analisadas 14,52%(n=27/186) foram consideradas positivas para hemoplasmas hemotrópicos através de PCR usando os primers Myco 322s e Myco 938as, que amplificam 620 pares de bases do fragmento de 16S rDNA. O diagnóstico para Anaplasmataceae foi dificultado pela inespecificidade das reações de PCR com os pares de primers testados, e quando a técnica foi específica, resultou em fragmento de 16S rDNA de Wolbachia sp. Por esse motivo, estes dados de positividade não foram considerados para análise. A positividade em lâmina de esfregaço sanguíneo para bactérias da família Anaplasmataceae foi de 6,99% (n=13/186), enquanto para micoplasmas hemotrópicos foi de 1,61% (n=3/186). A localidade de Santa Cecília apresentou a maior frequência de positividade para no diagnóstico molecular de Mycoplasma spp. (25,9%, n=7/48). Com relação ao resultado de sequenciamento do fragmento do 16S rDNA de micoplasmas hemotrópicos, três amostras de morcegos dos gêneros Glossophaga (ID 22052), Artibeus (ID 22108), Carollia (ID 22152) tiveram suas sequências com boa leitura no cromatograma, com aproximadamente 600pb. O maior percentual de similaridade Blast para 22052 foi de 98,41% com amostra de Mycoplasma sp. de um Glossophaga commissarisi (LR699022) da Costa Rica; para 22108 foi de 99,82% com amostra de Mycoplasma sp. de um Platyrrhinus lineatus (MZ048291) do Mato Grosso do Sul, Brasil; e para 22152 foi de 98,28% com amostra de Mycoplasma sp. de um Desmodus rotundus de Belize (KY932722). Este estudo revelou diferentes genótipos de micoplasmas hemotrópicos presentes em morcegos e demonstrou a presença de mórulas características de bactérias da família Anaplasmataceae em leucócitos e plaquetas de roedores, marsupiais e morcegos. Estes achados sugerem que micoplasmas hemotrópicos circulem entre morcegos do Acre, e que muito ainda há de ser descoberto em relação as espécies de bactérias intracelulares obrigatórias nestes animais. | pt_BR |

| dc.description.abstractOther | The aim of this study was to detect and molecularly characterize bacteria from the Anaplasmataceae and Mycoplasmataceae families in populations of small mammals in areas of the Amazon rainforest in the state of Acre, Brazil. The importance of this research is based on obligate intracellular bacteria that participate in wild cycles as zoonotic agents, and therefore of interest to human health, as well as parasites of importance to domestic animals, and the host animal's own ecology and life cycle. The study areas are concentrated in jungle areas of the Amazon rainforest, in the city of Rio Branco, state of Acre, Brazil. For the direct diagnosis, 186 samples of blood smear slides from previously captured rodents, marsupials and bats were used to detect and characterize bacteria from the Anaplasmataceae and Mycoplasmataceae families. Liver parenchyma samples stored in RNA Later® had their DNA extracted using the "DNeasy Blood & Tissue" kit (Qiagen®) and were used for molecular diagnosis. Of the samples analyzed, 14.52% (n=27/186) were positive for hemotropic hemoplasmas by PCR using the primers Myco 322s and Myco 938as, which amplify 620 base pairs of the 16S rDNA fragment. Diagnosis for Anaplasmataceae was hampered by the non-specificity of the PCR reactions with the primer pairs tested, and when the technique was specific, it resulted in a 16S rDNA fragment of Wolbachia sp. For this reason, these positivity data were not considered for analysis. Positivity on blood smear slides for bacteria from the Anaplasmataceae family was 6.99% (n=13/186), while for hemotropic mycoplasmas was 1.61% (n=3/186). Santa Cecília had the highest positive rate in molecular diagnosis of Mycoplasma spp. (25.9%, n=7/48). The results of sequencing of the 16S rDNA fragment of hemotropic mycoplasmas, three bat samples from the genera Glossophaga (ID 22052), Artibeus (ID 22108), Carollia (ID 22152) had their sequences read well on the chromatogram, with approximately 600pb. The highest percentage of Blast similarity for 22052 was 98.41% with a sample of Mycoplasma sp. from a Glossophaga commissarisi (LR699022) from Costa Rica; for 22108 it was 99.82% with a sample of Mycoplasma sp. from a Platyrrhinus lineatus (MZ048291) from Mato Grosso do Sul, Brazil; and for 22152 it was 98.28% with a Mycoplasma sp. sample from a Desmodus rotundus from Belize (KY932722). This study revealed different genotypes of hemotropic mycoplasmas present in bats and demonstrated the presence of morulae characteristic of bacteria from the Anaplasmataceae family in leukocytes and platelets from rodents, marsupials and bats. These findings suggest that hemotropic mycoplasmas circulate among bats in Acre, and that much remains to be discovered about the species of obligate intracellular bacteria in these animals. | en |
