| dc.description.abstract | A resistência à colistina representa uma grande ameaça à saúde global, comprometendo a eficácia de um dos antibióticos de último recurso para infecções causadas por Gram-negativos multirresistentes. Nos últimos anos, a detecção de variantes móveis dos genes mcr em ambientes associados à produção animal tem intensificado as preocupações quanto ao papel desses sistemas na manutenção e disseminação de determinantes de resistência. Este estudo teve como objetivo caracterizar Enterobacterales resistentes às polimixinas em águas residuais de um abatedouro avícola no município de São José do Vale do Rio Preto, RJ, Brasil. Entre setembro de 2021 e junho de 2022, foram realizadas quatro coletas em três pontos do sistema de tratamento (efluente bruto, tanque de floculação e efluente tratado). Um total de 140 cepas Gram-negativas foi recuperado, das quais 39 (27,8%) apresentaram resistência à colistina (CIM ≥ 2 μg/mL). A análise molecular revelou a presença de genes mcr-1 e mcr-10 em 41% (16/39) dessas cepas. O gene mcr-1 foi detectado em E. coli, K. pneumoniae e Enterobacter spp., enquanto o mcr-10 foi identificado exclusivamente em membros do complexo Enterobacter cloacae. As análises de MLST evidenciaram a ocorrência de clones de importância clínica como K. pneumoniae ST15 e novos clones de interesse Ent. bugandensis ST3343, ambos recuperados em todos os pontos do sistema, inclusive no efluente tratado. Entre as cepas de E. coli, foram identificados sete diferentes STs, revelando a diversidade clonal e a persistência de cepas ao longo do tratamento. A análise filogenética pelo método de Clermont indicou que todas as cepas pertencem ao grupo E. O sequenciamento genômico completo mostrou a presença do plasmídeo IncX4 carregando mcr-1 em K. pneumoniae ST15, além de múltiplos genes de resistência. Já a cepa de Enterobacter pertencente ao complexo Enterobacter cloacae foi identificada como sendo da espécie vonholyi, de acordo com a análise no TYGS, e apresentou mcr-10 em associação com outros determinantes como blaACT, oqxAB e fosA. Este achado representa, à luz das evidências publicadas, o primeiro relato da presença de mcr-10 em Enterobacter spp. no Brasil. Essas descobertas evidenciam que águas residuais de abatedouro avícola podem atuar como importantes reservatórios ambientais de genes de resistência, destacando a necessidade de vigilância integrada e regulamentação mais rigorosa do uso de colistina na produção animal, para mitigar a disseminação ambiental da resistência a antibióticos de último recurso. | pt_BR |

| dc.description.abstractOther | Colistin resistance represents a major threat to global health, compromising the efficacy of one of the last-resort antibiotics for infections caused by multidrug-resistant Gram-negative bacteria. In recent years, the detection of mobile mcr gene variants in environments associated with animal production has raised concerns about their role in the maintenance and dissemination of resistance determinants. This study aimed to characterize Enterobacterales resistant to polymyxins in wastewater from a poultry slaughterhouse located in São José do Vale do Rio Preto, Rio de Janeiro, Brazil. Between September 2021 and June 2022, four sampling campaigns were conducted at three points of the treatment system (raw effluent, flocculation tank, and treated effluent). A total of 140 Gram-negative isolates were recovered, of which 39 (27.8%) showed resistance to colistin (MIC ≥ 2 μg/mL). Molecular analysis revealed the presence of mcr-1 and mcr-10 genes in 41% (16/39) of these isolates. The mcr-1 gene was detected in Escherichia coli, Klebsiella pneumoniae, and Enterobacter spp., whereas mcr-10 was identified exclusively in members of the Enterobacter cloacae complex. MLST analysis revealed the occurrence of clinically relevant clones such as K. pneumoniae ST15 and new clones of interest, including Enterobacter bugandensis ST3343, both recovered from all sampling points, including the treated effluent. Among E. coli isolates, seven distinct sequence types (STs) were identified, demonstrating clonal diversity and persistence throughout the treatment process. Phylogenetic analysis using the Clermont method showed that all E. coli strains belonged to phylogroup E. Whole-genome sequencing revealed the presence of an IncX4 plasmid carrying mcr-1 in K. pneumoniae ST15, along with multiple antimicrobial resistance genes. The Enterobacter isolate belonging to the E. cloacae complex was identified as E. vonholyi by TYGS analysis and harbored mcr-10 in association with other resistance determinants, including blaACT, oqxAB, and fosA. To the best of our knowledge, this represents the first report of mcr-10 in Enterobacter spp. in Brazil. These findings highlight that poultry slaughterhouse wastewater can serve as an important environmental reservoir of resistance genes, underscoring the need for integrated surveillance and stricter regulation of colistin use in animal production to mitigate the environmental dissemination of resistance to last-resort antibiotics. | en |
