dc.description.abstractA menor fertilidade do sêmen sexado tem sido atribuída a fatores como a concentração espermática e danos causados durante o processo de sexagem, reduzindo sua viabilidade no sistema reprodutivo das fêmeas. Sendo assim, o momento da inseminação em relação à ovulação é de fundamental importância para melhorar a fertilidade da técnica. Portanto, a administração de análogos do GnRH, como o acetato de buserelina (AB), no momento da inseminação, pode se tornar uma estratégia interessante no intuito de antecipar a ovulação. Assim, a hipótese do presente estudo é que a aplicação de AB no momento da inseminação aumenta a taxa de concepção de vacas leiteiras inseminadas com sêmen sexado. Neste contexto, o objetivo geral do presente estudo foi comparar as taxas de concepção de fêmeas leiteiras inseminadas com sêmen sexado após a aplicação ou não de AB. Adicionalmente, foram avaliados os efeitos da ciclicidade das fêmeas no início da sincronização e do diâmetro do folículo pré-ovulatório (FPO) nas taxas de concepção. Para tanto, 116 fêmeas bovinas mestiças (Girolando) provenientes de fazendas leiteiras comerciais foram submetidas a um protocolo de sincronização da ovulação. As fêmeas foram divididas aleatoriamente em dois grupos: Grupo acetato de buserelina (GB; n=58), que recebeu 10μg de AB no momento da inseminação e Grupo Controle (GC; n=58), que não recebeu qualquer hormônio no momento da inseminação. O protocolo de sincronização utilizado incluiu a inserção de um dispositivo intravaginal de progesterona associado a 2mg de benzoato de estradiol em dia aleatório do ciclo estral (D0). Nove dias depois (D9), o dispositivo foi removido e foram aplicadas 300UI de gonadotrofina coriônica equina + 1mg de cipionato de estradiol + 500μg de cloprostenol sódico (prostaglandina). A inseminação foi realizada com sêmen sexado e ocorreu 48 horas após a remoção do dispositivo de progesterona (D11). No D0 e D11 as fêmeas foram submetidas à avaliação ultrassonográfica para determinação da ciclicidade e mensuração do diâmetro do FPO. O diagnóstico de gestação foi realizado entre 30 e 40 dias após a inseminação. As variáveis concepção, tratamentos, ciclicidade, ECC (Escore de Condição Corporal), categoria, fazenda, touro e interações foram avaliadas por regressão logística multivariada utilizando o programa Bioestat 5.3. A variável diâmetro do FPO foi primeiro analisada para heterogeneidade de variância e normalidade e posteriormente comparada por ANOVA. Não foi observada diferença significativa (P = 0,13) entre as taxas de concepção dos grupos GB e GC (22,4% e 13,8%, respectivamente). No entanto, quando comparou-se as taxas de concepção apenas dos animais que estavam em anestro ao início do tratamento, houve tendência (P=0,07) da taxa de concepção ser maior nos animais do GB. Quando as taxas de concepção foram comparadas de acordo com o diâmetro do FPO, observou-se que animais do GB com FPO <13mm apresentaram maiores taxas de concepção em relação aqueles com FPO ≥13mm (P=0,01). Portanto, conclui-se que o acetato de buserelina administrado no momento da inseminação em fêmeas bovinas inseminadas com sêmen sexado não melhora a taxa de concepção, exceto quando estas apresentam FPO <13mm.pt_BR

dc.description.abstractOtherThe lower fertility of sexed semen has been attributed to factors such as sperm concentration and damage caused during the sexing process, reducing its viability in the female reproductive system. Therefore, the timing of insemination in relation to ovulation is of fundamental importance to improve the fertility of the technique. Therefore, the administration of GnRH analogues, such as buserelin acetate (BA), at the time of insemination may become an interesting strategy to anticipate ovulation. Thus, the hypothesis of the present study is that the application of BA at the time of insemination increases the conception rate of dairy cows inseminated with sexed semen. In this context, the general objective of the present study was to compare the conception rates of dairy cows inseminated with sexed semen after the application or not of BA. Additionally, the effects of female cyclicity at the beginning of synchronization and preovulatory follicle diameter (POF) on conception rates were evaluated. For this purpose, 116 crossbred female cattle (Girolando) from commercial dairy farms were subjected to an ovulation synchronization protocol. The females were randomly divided into two groups: Buserelin acetate group (GB; n=58), which received 10μg of AB at the time of insemination, and Control group (CG; n=58), which did not receive any hormone at the time of insemination. The synchronization protocol used included the insertion of an intravaginal progesterone device associated with 2mg of estradiol benzoate on a random day of the estrous cycle (D0). Nine days later (D9), the device was removed and 300IU of equine chorionic gonadotropin + 1mg of estradiol cypionate + 500μg of sodium cloprostenol (prostaglandin) were applied. Insemination was performed with sexed semen and occurred 48 hours after removal of the progesterone device (D11). On D0 and D11, the females underwent ultrasonographic evaluation to determine cyclicity and measure the diameter of the POF. Pregnancy diagnosis was performed between 30 and 40 days after insemination. The variables conception, treatments, cyclicity, BCS (Body Condition Score), category, farm, bull and interactions were evaluated by multivariate logistic regression using the Bioestat 5.3 program. The variable POF diameter was first analyzed for heterogeneity of variance and normality and subsequently compared by ANOVA. No significant difference (P = 0.13) was observed between the conception rates of the GB and GC groups (22.4% and 13.8%, respectively). However, when comparing the conception rates of animals that were in anestrus at the beginning of treatment, there was a tendency (P=0.07) for the conception rate to be higher in GB animals. When the conception rates were compared according to the POF diameter, it was observed that GB animals with POF <13 mm had higher conception rates compared to those with POF ≥13 mm (P=0.01). Therefore, it is concluded that buserelin acetate administered at the time of insemination in female cattle inseminated with sexed semen does not improve the conception rate, except when these animals have POF <13 mm.en
