dc.description.abstractNeste estudo testamos a atividade anti-Leishmania dos óleos essenciais de Lippia sidoides Cham. e seu composto majoritário timol e Aloysia gratíssima (sin. Lippia lycioides) e seu composto majoritário guaiol, utilizando Leishmania amazonensis como modelo experimental in vitro. Nossos resultados demonstraram que os óleos essenciais (OEs) de L. sidoides Cham. e A. gratíssima (sin. L. lycioides), apresentaram efeito em promastigotas, com IC50 (48h) 22,2 ug/mL e 25,5 ug/mL, IC50 (72h) 4,4 ug/mL e 14,2 ug/mL, respectivamente. Ambos OEs também apresentaram efeito em formas amastigotas, onde o OE de A. gratíssima (sin. L. lycioides) com IC50 1 ug/mL de forma dose-dependente, e os compostos timol e guaiol também apresentaram efeito em amastigotas, onde o guaiol apresentou um IC50 0,7 ug/mL, igual ao óleo essencial A.gratissima (sin. L. lycioides), e as concentrações testadas de timol não chegaram a concentração de 50 % de inibição. A citotoxicidade destes componentes para os macrófagos peritoneais murinos foi avaliada através do método de XTT onde não apresentaram atividade mitocondrial alterada, a integridade da membrana dos macrófagos também não foi alterada após o tratamento. Os dois óleos essenciais testados não foram capazes de alterar a capacidade fagocítica da célula hospedeira. O efeito anti-Leishmania amazonensis dos óleos essenciais de L. sidoides Cham. e A.gratissima (sin. L. lycioides), testados é independente da produção de óxido nítrico. Nossos resultados apontam os óleos essenciais de L. sidoides Cham. e A.gratissima (sin. L. lycioides), além de seus componestes majoritários timol e guaiol, respectivamente, com potencial para o desenvolvimento de terapia contra as leishmanioses.pt_BR

dc.description.abstractOtherIn this study we tested the anti-Leishmania activity of essential oils from Lippia sidoides Cham. and its major compound thymol and Aloysia gratissima (syn. Lippia lycioides) and its major compound guaiol using Leishmania amazonensis as an experimental model in vitro. Our results demonstrated that essential oils (EOs) of L. sidoides Cham. and Aloysia gratissima (syn. L. lycioides) showed effect in promastigotes, with IC50 (48h) 22.2 ug/mL and 25.5 ug/mL, IC50 (72h) 4.4 ug/mL and 14.2 ug/mL, respectively. Both EOs also had effect on amastigotes, where the EO of A. gratissima (syn. L. lycioides) with IC50 1 ug/mL dose-dependent manner, and the compounds thymol and guaiol also showed effect in amastigotes, where guaiol showed an IC50 of 0.7 ug/mL equal to essential oil of A. gratissima (syn. L. lycioides) and thymol the tested concentrations did not reach concentrations of 50% inhibition. Cytotoxicity of these components for murine peritoneal macrophages was assessed by the XTT method which did not show altered mitochondrial activity, the macrophage membrane integrity was not altered after the treatment. The two essential oils tested were not able to alter the phagocytic ability of the host cell. The anti-Leishmania amazonensis essential oils of L. sidoides Cham. and A.gratissima (syn. L. lycioides) tested is independent of nitric oxide production. Our results indicate the essential oils of L. sidoides Cham. and A. gratissima (syn. L. lycioides), and its majority compound guaiol and thymol, respectively, with potential for the development of therapy against leishmaniasis.en
