| dc.description.abstract | Foi realizada a caracterização morfológica de isolados de rizóbio provenientes de nódulos de caule e raiz de Discolobium pulchellum e Discolobium psoraleaefolium, leguminosas nativas de áreas alagadas do Pantanal Mato- Grossense, e a avaliação da tolerância destes isolados a níveis crescentes de NaCl e altas temperaturas. Foram caracterizados 13 isolados de nódulos de caule e 14 isolados de nódulos de raiz. Dentre os isolados de nódulos de raiz de Discolobium spp., 90% apresentaram crescimento lento alcalinizando o meio, ao passo que os isolados de nódulos de caule, 46% apresentaram crescimento lento alcalinizando o meio. Os isolados de crescimento rápido mostraram-se mais tolerantes às crescentes concentrações salinas do que os de crescimento lento e também foram mais tolerantes ao estresse de temperaturas elevadas em relação aos isolados de crescimento lento. Isolados selecionados aleatoriamente dentro de cada grupo formado a partir da caracterização morfológica, foram testados quanto à capacidade de nodulação em caule e raiz e potencial de fixação biológica de nitrogênio em Discolobium pulchellum sob condições de inundação. Os resultados obtidos mostraram não haver especificidade de nodulação dos isolados testados provenientes de nódulos de caule e de raiz. Em relação ao potencial de fixação biológica de nitrogênio de D. pulchellum, os resultados mostraram que os isolados de crescimento lento que alcalinizam o meio YMA formaram simbiose eficiente, e que os isolados de crescimento rápido produzindo ácido no mesmo meio formaram nódulos ineficientes. Pode-se observar a formação do nódulo em caule de Discolobium pulchellum 12 dias após a inoculação. Observou-se a atividade mitótica de células do parênquima cortical durante a formação do primórdio nodular com contínuo aumento do tamanho do nódulo e colonização das células da zona meristemática pela bactéria. Após 4 dias do início de formação do nódulo pode-se observar uma abundante colonização intracelular da bactéria. Nódulos funcionais de D. pulchellum foram observados aos 20 dias após a inoculação, onde pode-se observar que não havia células intersticiais não infectadas no tecido infectado. Foi observada uma nova espécie dentro do gênero Discolobium, Discolobium leptophyllum, com capacidade de formar nódulos no caule em condições de inundação. Os nódulos de caule são conectados ao caule e o sistema vascular do caule penetra no nódulo. Os nódulos são do tipo aesquinomenóide desmodióide, ou seja, sem células não infectadas no tecido infectado. | pt_BR |

| dc.description.abstractOther | Forteen root and thirteen stem nodules isolates from Discolobium pulchellum and D. psoraleaefolium were characterized in Yeast Manitol Agar (YMA) culture medium and tested for tolerance and salinity and high temperature. Ninety percent of the root nodule isolates were slow growth alkali producing and forty six percent of stem nodule isolates were slow growth alkali producing. The fast growing isolates were more tolerant to salinity and to high temperature. Representative isolates from root and stem nodules of each group were tested for cross inoculation of root and stems of D. pulchellum growing under flooded conditions. Both groups, root and stem isolates, nodulated root and stem. Slow growing alkali producing isolates formed efficient symbiosis with D. pulchellum, while fast growing acid producing, produced inefficient nodules. Stem nodules of D. pulchellum were formed 12 days after inoculation. Mitotic activity of the cortical parenchyma was present at nodule primordia formation with a continuous growth and meristematic cells colonization by the bacteria. After 4 days of nodule initiation it was observed a profuse intracellular colonization. Mature nodules of D. pulchellum were observed 20 days after inoculation without uninffected cells infected tissue. A new Discolobium species, D. leptohyllum was observed to nodulate root and also stems when flooded. As observed for D. pulchellum, the new species also presented nodules with vascular bundle directly connected to the stem vascular bundle. The nodules of D. leptophyllum are also aeschinomenoid desmodioid type, without uninfected cells in the infected tissue. | en |
