In a first phase, the effect of pre-cropping with legumes and sorghum on the inoculum potential of arbuscular mycorrhizal fungi and cassava production was evaluated. The experiment consisted of two cultivation cycles. The first involved planting different plant covers and a treatment with no cover, and the second involved planting cassava. The experiment was set up at CNPAB-EMBRAPA - Seropédica, RJ, under field conditions, in 30 m² plots, in a randomized complete block design with six treatments and four replications. As pre-crops (first cycle), four legume species and one grass were evaluated, namely: jack bean, velvet bean, sunn hemp, pigeon pea, and sorghum (C.V. BR 005), in addition to the bare soil treatment (soil kept without plants). The plants grew for 82 days. The second cycle began after cutting the shoots of the cover crops, at which point cassava cv. Saracura was planted. Cassava root yield ranged from 12.76 to 17.20 t ha⁻¹; however, the treatments did not produce significant differences.Sorghum plants increased the number of infective propagules compared to the bare soil, demonstrating that the use of plant species with the capacity to increase the number of infective propagules is a viable alternative to the problem of inoculating mycotrophic crops at the field level. The data showed that the Most Probable Number technique, although laborious, is sensitive enough to differentiate the effect of treatments compared to the spore counting technique. Sixteen species of AM fungi were identified, and it was found that some species underwent a change in their frequency of occurrence with cultivation.Subsequently, the response to inoculation with the fungus Glomus manihotis on cassava production was evaluated under greenhouse conditions in a randomized complete block design with 5 replications, in a 3x3x2 factorial scheme, namely: 3 soil samples collected under different plant covers (bare soil, sunn hemp, and sorghum); 3 inoculation sources (absence, spore suspension, and soil inoculum); and two soil disinfestation levels (absence and presence of fumigation).Pre-cropping with sunn hemp provided the greatest benefits, stimulating mycorrhization in the non-fumigated soil and promoting increases in N accumulation and total dry matter of both roots and shoots in both soil disinfestation treatments compared to the other pre-crops. Inoculation promoted an increase in cassava shoot dry matter production compared to the uninoculated treatment in the fumigated soil and increased the accumulation of N, P, and K. Furthermore, pre-cropping with sunn hemp inoculated with soil inoculum promoted greater N accumulation compared to the other pre-crops. In the non-fumigated soil, there was no response to inoculation. Cassava shoot dry matter production in the non-fumigated soil was, on average, 42.7% lower than in the fumigated soil. Conversely, root production showed the opposite behavior, yielding 41.9% more compared to the fumigated soil.
