dc.description.abstractAs leishmanioses são um grupo de doenças causadas por protozoários do gênero Leishmania, com uma vasta distribuição mundial e de alta relevância em saúde pública, ainda assim é negligenciada, acometendo principalmente países ainda em desenvolvimento. O Brasil mantém a maior parte dos registros de casos por leishmaniose da América Latina, apresentando formas tegumentares e viscerais da doença. Sua transmissão ocorre através de flebotomíneos infectados e possuem múltiplos reservatórios, em especial os canídeos. Contudo, a leishmaniose felina (FeL) tem sido cada vez mais descrita na literatura, o que sugere um potencial papel dos gatos como reservatórios secundários, principalmente, em áreas endêmicas. Desta forma, este trabalho tem o objetivo de estabelecer um modelo de infecção felina in vitro para elucidar os aspectos biológicos e imunológicos da leishmaniose felina, buscando caracterizar a linhagem de macrófagos felinos Fcwf-4 durante a infecção por Leishmania amazonensis e promover um modelo in vitro para futuros estudos. Foi observado que essa célula é capaz de promover interação com o parasita e que estes permanecem viáveis por pelo menos 48 horas. A fim de simular uma coinfecção, foi utilizado o polissacarídeo capsular glucuronoxilomanana (GXM) de Cryptococcus neoformans,que pode imunomodular negativamente a resposta celular, nossos dados mostraram que na presença deste, há alteração na produção de óxido nítrico (NO) e radicais de oxigênio (ROS), e na expressão de citocinas, o que favorece permanência intracelular do parasitapt_BR

dc.description.abstractOtherLeishmaniases are a group of diseases caused by protozoa of the genus Leishmania, with a wide worldwide distribution and high relevance to public health; nevertheless, they remain neglected and mainly affect developing countries. Brazil accounts for most of the reported cases of leishmaniasis in Latin America, presenting both cutaneous and visceral forms of the disease. Transmission occurs through infected sandflies and involves multiple reservoirs, especially canids. However, feline leishmaniasis (FeL) has been increasingly reported in the literature, suggesting a potential role of cats as secondary reservoirs, particularly in endemic areas. Therefore, this study aimed to establish an in vitro feline infection model to elucidate the biological and immunological aspects of feline leishmaniasis, seeking to characterize the feline macrophage cell line Fcwf-4 during infection with Leishmania amazonensis and to provide an in vitro model for future studies. It was observed that this cell line is capable of interacting with the parasite and that the parasites remain viable for at least 48 hours. In order to simulate a coinfection, the capsular polysaccharide glucuronoxylomannan (GXM) from Cryptococcus neoformans, which can negatively immunomodulate the cellular response, was used. Our data showed that in its presence there are changes in nitric oxide (NO) and reactive oxygen species (ROS) production, as well as in cytokine expression, which favors the intracellular persistence of the parasiteen
