dc.description.abstractNo presente trabalho, taquizoítas do Toxoplasma gondii isolados de cérebro e coração de uma galinha naturalmente infectada, procedente de área urbana do município de Seropédica, Estado do Rio de Janeiro, e mantidos em camundongos albinos foram estudados e comparados à cepa congénita, oriunda do IOC-FIOCRUZ, Rio de Janeiro. As amostras foram submetidas a eletroforese em gel de SDS- poliacrilamida a fim de se avaliar as possíveis variações protéicas em cada uma das amostras. Após montagem dos géis, integrou-se os zimogramas por densitometria a laser, onde se analisou as áreas (concentrações) de proteínas em cada amostra. Os perfis protéicos eram distintos mesmo quando comparados à cepa “C”, evidenciando-se com isso, variações intraespecíficas observadas em uma espécie hospedeira. Da mesma maneira, ao se analisar os zimogramas, observou-se que haviam percentuais distintos nas concentrações protéicas entre as amostras cérebro e coração e essas com a cepa “C” do parasito. Para avaliar-se o reconhecimento de determinantes antigénicos do material estudado, procedeu-se o ensaio imunoenzimático “Western-Blotting” frente à soro hiperimune de coelho e soro de cão positivo na IF e soros negativos, respectivamente, para T. gondii. As análises das fitas antigênicas, frente ao soro de coelho, revelaram reconhecimentos distintos entre as amostras de T. gondii isolado de Gallus gallus domesticus naturalmente infectado e dessas com a cepa “C”, embora o grupo compartilhe de cinco bandas anti-IgG para este hospedeiro (P30; P34, 7; P43; P67 e P80). Já frente ao soro de cão, observou-se o reconhecimento de dois determinantes antigénicos idênticos (P30 e P34,7) para as amostras de cérebro e coração. Esses e outros determinantes distintos foram reconhecidos nesse hospedeiro para a cepa “C”.pt_BR

dc.description.abstractOtherIn the present study, taquizoites of Toxoplasma gondii isolated from naturally infected Gallus gallus domesticus brain and heart, from the urban area of Seropedica municipality, Rio de Janeiro State, and maintained in albine mice were studied and compared with the congenital strain, from IOC-FIOCRUZ, Rio de Janeiro. Strains were underwent to eletrophoresis SDS-polyacrylamide gel in order to evaluate possibles protein variations in each strain. After the make-up of the gel, zimograme was binded by laser densitometry, where protein areas (concentration) in each strain were analised. Protein outlines were distinct, even when compared to strain “C”, showing intra-specific variations observed in the same host species. in the same way, analyzing zimogrames, distinct percentuals were observed in protein concentration between brain and heart strains and between these strains and parasite strain “C”. Evaluating the recognition of studied material antigenic determinants, the enzime imunoassay Western-blotting was applied to rabbit hiperimune serum and enzime imunoassay Western-blotting was applied to rabbit hiperimune serum and dog serum, both positives, at it and negative serum respectively. Antigenic strip analysis of the rabbit serum, revealed distincts recognitions between T. gondii strains isolated from naturally infected chicken and between these strains and strain “C”, although the group shares five bands anti-igG for this host (P30; P34,7; P43; P67 and P80). The recognition of two equal antigenic determinants (P30 and P34,7) in the dog serum for brain and heart strains was observed. These and others distincts determinants were recognized in this host for strain “C”.en
