| dc.description.abstract | A fixação biológica de nitrogênio (FBN) em plantas de cana-de-açúcar tem sido considerada um dos principais suportes para o PROÁLCOOL, influindo de maneira positiva para o aumento do balanço energético na produção de álcool combustível. Sendo a FBN resultado de complexas reações fisiológicas e bioquímicas, esta é dependente da expressão do potencial genético do microrganismo diazotrófico e do hospedeiro, podendo ser limitada por diversos fatores, tanto bióticos, quanto abióticos. Neste estudo buscou-se avaliar o efeito de diferentes fatores sobre a presença e/ou o número de bactérias diazotróficas presentes na cultura da cana-de-açúcar. A influência do genótipo da planta foi estudada fazendo-se um levantamento das bactérias diazotróficas presentes, e também com a quantificação da população de Herbaspirillum spp. e A. diazotrophicus, em quatro genótipos de cana-de-açúcar contrastantes quanto a capacidade de obter nitrogênio via FBN. Os genótipos avaliados neste trabalho foram Krakatau, CB 45-3, SP 70-1143, ditas eficientes em relação a FBN e Chunee citada como sendo um genótipo pouco eficiente. De acordo com o levantamento realizado as bactérias estudadas (Azospirillum lipoferum, A. brasilense, A. amazonense, Herbaspirillum spp. e Acetobacter diazotrophicus) estavam presentes em todos os genótipos avaliados. Já a quantificação das bactérias Herbaspirillum spp. e A. diazotrophicus mostrou que não há diferenças significativas entre os genótipos quanto ao número destas bactérias. Também foi observado que enquanto Herbaspirilllum spp. mantem-se mais estável ao longo do ciclo da cultura, a população de A. diazotrophicus parece sofrer um decréscimo com a aproximação do final do ciclo comercial. Estes resultados podem sugerir que as diferenças entre as taxas de FBN encontradas nos diversos genótipos não são causadas por diferenças na presença ou número das espécies de bactérias aqui estudadas. Em outro estudo buscou-se evidências da presença destas bactérias em várias amostras de plantas micropropagadas, desde a fase inicial do processo de micropropagação até a utilização destas plantas no campo. Os estudo de ocorrência destas bactérias nestes materiais mostrou que o cultivo de meristemas realmente elimina as bactérias diazotróficas endofíticas, já que estas não foram encontradas no material recém micropropagado. No entanto, foi possível encontrar Azospirillum spp. em plantas ainda na fase de aclimatação, e as bactérias Herbaspirillum spp. e Acetobacter diazotrophicus foram isoladas de plantas que já vinham sendo utilizadas no campo. Também foram testadas duas novas alternativas de isolamento para Herbaspirillum spp. e A. diazotrophicus, e tanto a pré- germinação de toletes colhidos no campo, quanto a técnica de imunocaptura mostraram-se alternativas viáveis para o isolamento destas bactérias. Os estudos de microscopia com plântulas micropropagadas inoculadas com a estirpe PAL 5 de A. diazotrophicus, mostraram aos 7 dias após a inoculação, que as bactérias estavam localizadas preferencialmente em feridas naturais ou naquelas provocadas pela emergência de raízes laterais, como também na ponta das raízes (região meristemática), provavelmente utilizando estas regiões para atingir o interior da planta. Já no interior das raízes pode-se observar as bactérias colonizando o espaço intercelular da região do córtex, como também a presença de células nos vasos do xilema, provavelmente fazendo desta região uma via de acesso a parte aérea das plantas. Por fim, foram implantados dois experimentos para averiguar os possíveis efeitos da adubação com altos níveis de N, em plantas de cana-de- açúcar, sobre as bactérias diazotróficas Herbaspirillum spp. e Acetobacter diazotrophicus. Em um experimento de campo obeservou-se o comportamento da população natural destas bactérias, utilizando-se plantas de cana-de-açúcar adubadas com doses equivalentes a 300 kg de N/ha. Na genótipo SP 79-2312, de maneira geral, em comparação com plantas onde não foi feita a aplicação de N, as plantas adubadas mostraram maior concentração deste elemento nos tecidos das raízes e colmos. Estas mesmas plantas também apresentaram um número menor da bactéria A. diazotrophicus, enquanto a população de Herbaspirillum spp. não foi afetada. A população de A. diazotrophicus também mostrou ser influenciada pela época de coleta, apresentando-se reduzida nas coletas coincidentes com os períodos secos do ano, e maior nos períodos de maior intensidade pluviométrica. Em um segundo experimento, montado em casa de vegetação, utilizou-se plântulas micropropagadas da genótipo Na 56-79, inoculadas com estirpes de A. diazotrophicus e H. seropedicae. Não foi demonstrado efeito da adubação nitrogenada sobre a população de ambas bactérias. A adubação nitrogenada foi prejudicial ao crescimento radicular, e a inoculação com estas bactérias, principalmente H. seropedicae, diminuiu o efeito deletério causado pela adição de N. Este efeito da inoculação apresentado nas raízes, provavelmente também foi o responsável por um maior desenvolvimento inicial da parte aérea nas plantas inoculadas e adubadas. | pt_BR |

| dc.description.abstractOther | The biological nitrogen fixation (BNF) in sugar-cane plants has been considered one of the main supports for the PROÁLCOOL programme, influencing in a positive way the increase of the energy balance for the production of alcohol combustible. BNF is result of complex physiological and biochemical reactions, and is dependent of the genetic potential expression of the diazotroph microrganism and the plant. It could be limited by several factors, biotic and abiotic. In this study was evaluated the effect of different factors on the presence and/or the number of the diazotrophic bacteria present in the sugar-cane plants. The influence of the plant genotype was studied performing a survey of the diazotrophic bacteria present in four sugar-cane genotypes differing in their capacity to obtain nitrogen through BNF, and also quantified the population of Herbaspirillum spp. and A. diazotrophicus within them. The genotypes evaluated were Krakatau, CB 45-3, SP 70-1143, all of which have been shown to be efficient in relation to BNF, and Chunee, a non- efficient genotype. The bacteria tested in the survey were Azospirillum lipoferum, A. brasilense, A. amazonense, Herbaspirillum spp. and Acetobacter diazotrophicus. All these bacteria were present in the four examined genotypes. The quantification of Herbaspirillum spp. and A. diazotrophicus showed that there were no significant differences among the sugar cane genotypes with respect to the number of these bacteria. While numbers of Herbaspirilllum spp. remained stable during the life-cycle of the culture, the population of A. diazotrophicus seemed to suffer a decrease with the approach of the end of the commercial cycle. Taken together, these results suggest that the differences in the rates of BNF found in sugar cane genotypes are not caused by differences in the presence or the number of the bacterial species studied here. In another study, evidence of the presence of some of these bacteria were looked for in several samples of micropropagated plants, from the initial phase of the micropropagation process until being planted in the field. The study demonstrated that the tissue culture technique does indeed eliminate the endophytic diazotrophic bacteria, as none were found in the newly micropropagated material. However, it was possible to find Azospirillum spp. in plants in the acclimatization phase, while isolates of Herbaspirillum spp. and A. diazotrophicus were obtained from plants that had already became used to the field. Also in this chapter, two new isolation alternatives for Herbaspirillum spp. and A. diazotrophicus were tested, and it was shown that pre-germination of stems picked in the field, together with the imunotrapping technique, were viable alternatives for the isolation of these bacteria. Microscopic studies were also made with micropropagated sugar-cane plantlets inoculated with the PAL 5 strain of A. diazotrophicus. These showed that 7 days after inoculation, the bacteria were located mainly within natural wounds, or within those “wounds” produced during the emergence of lateral roots, as well as in the root tip (meristematic area). These regions were probably used by the bacteria to enter the plants. Once inside the roots the bacteria were observed colonizing the intercellular spaces of the cortex, as well as the xylem vessels, probably using the latter as an access road to the aerial parts of the plants. Finally, two experiments were planted to study the possible effects of fertilization with high levels of N on the occurrence and numbers of the diazotrophic bacteria Herbaspirillum spp. and Acetobacter diazotrophicus in sugar-cane plants. In a field experiment the behaviour of the natural population of these bacteria was studied using plants fertilized with levels of up to 300 kg of N/ha. In the variety SP 79-2312, generally, in comparison to plants where no application of N was made, the fertilized plants showed larger concentrations of this element in the roots and stems. These same plants also had a smaller number of A. diazotrophicus, while the population of Herbaspirillum spp. was not affected by N application. The numbers of A. diazotrophicus were also shown to be influenced by the harvest time, becoming reduced in the harvests which coincided with dry periods of the year, and larger in the periods of larger pluviometric intensity. The second experiment was of a shorter duration (40 days) and was set up in a greenhouse. In this case micropropagated plantlets were inoculated with strains of A. diazotrophicus and H. seropedicae and no effects of nitrogen fertilization on the population of either bacteria were shown. However, it was demonstrated that nitrogen fertilization was harmful to root growth, and that inoculation with these bacteria, particularly H. seropedicae, decreased the deleterious effect caused by the addition of nitrogen. The inoculation effect shown by the roots was probably also responsible for a larger initial development of the aerial parts of the inoculated and fertilized plants. | en |
