dc.description.abstractO impacto ambiental derivado do uso excessivo de insumos químicos na agricultura convencional alerta para a necessidade de alternativas sustentáveis, com destaque para o aproveitamento biotecnológico da interação planta-microrganismo. Compreender a estrutura comunitária da microbiota endofítica, assim como os fatores moduladores da sua composição em agroecossistemas representa o passo inicial para a seleção e desenvolvimento de inoculantes. Desta forma, o presente trabalho teve como objetivo geral avaliar a composição de espécies das comunidades de fungos endofíticos septados escuros (Dark Septate Endophytes – DSE), fungos endofíticos radiculares no geral (FER) e fungos micorrízicos arbusculares (FMAs) associados à cana-de-açúcar no estado de São Paulo, Brasil, verificando possíveis relações entre a ocorrência das espécies e as características químicas do solo. Adicionalmente, o trabalho visou explorar o potencial de alguns isolados de DSE na promoção de crescimento vegetal. A simbiose com DSE foi observada nas variedades de cana RB867515, RB966928 e RB92579 e em uma variedade ainda não comercializada de cana-energia. A partir destas variedades, 16 fungos DSE foram isolados e identificados morfológica e molecularmente como pertencentes às ordens Helotiales, Pleosporales e Xylariales. Alguns destes fungos, com ênfase nos isolados de Periconia macrospinosa (Pleosporales), estimularam a absorção de P em plantas de cana e arroz, e solubilizaram óxido de zinco in vitro. A comunidade de FER associada à variedade de cana RB966928 em 24 plantações de São Paulo distribuídas na maioria das mesorregiões produtoras de cana-de-açúcar no estado foi dominada por espécies dos gêneros Fusarium e Trichoderma, assim como pela espécie de DSE P. macrospinosa. Já a comunidade de FMAs associada a esta variedade foi dominada pelas espécies Acaulospora mellea, A. scrobiculata, Glomus macrocarpum, G. microaggregatum, Racocetra persica e Sieverdingia tortuosa. Conclui-se que as simbioses com DSE e FMAs ocorrem e coexistem naturalmente nas raízes da cana-de-açúcar, independentemente da variedade. A análise de redundância (RDA) mostrou que a ocorrência de espécies de FMAs foi modulada pelas características químicas do solo, não sendo este o padrão verificado para a comunidade de FER.por

dc.description.abstractOtherEnvironmental problems derived from the excessive use of chemical inputs in conventional agriculture alerts towards the application of sustainable alternatives, with emphasis on the biotechnological use of the plant – microorganism interaction. Understanding the community structure of the endophytic microbiota, as well as the factors that modulate its composition in agroecosystems represent the initial step for the selection and developmnt of inoculants. The present work aimed to evaluate the diversity of dark septate endophytes (DSE), general root endophytic fungi (REF) and arbuscular mycorrhizal fungi (AMF) associated to sugarcane in the Brazilian state of São Paulo; searching for possible relationships between the occurrence of the fungal species and the soil chemical properties. Also, the potential of some DSE fungi in promoting plant growth was explored. Symbiosis with DSE was observed in the sugarcane varieties RB867515, RB966928 and RB92579 and in one not commercialized energy-cane variety. From the latter, 16 DSE fungi were isolated and identified within the orders Helotiales, Pleosporales and Xylariales. Some of these fungi, especially isolates of Periconia macrospinosa (Pleosporales), stimulated P uptake in sugarcane and rice plants, and solubilized zinc oxide in vitro. The REF community of the sugarcane variety RB966928, considering plants sampled in 24 fields of São Paulo, distributed among most of the mesoregions of the State, was dominated by species of the genera Fusarium and Trichoderma, as well as by the DSE P. macrospinosa. The AMF community associated with this sugarcane variety was dominated by the species Acaulospora mellea, A. scrobiculata, Glomus macrocarpum, G. microaggregatum, Racocetra persica e Sieverdingia tortuosa. It was concluded that symbioses with DSE and AMF occur and coexist naturally in the roots of sugarcane, regardless of the variety. As verified by the redundancy analysis (RDA), the occurrence of AMF species was modulated by the chemical characteristics of the soil, which was not the pattern verified for the REF community.eng
