dc.description.abstract | Plecoptera é uma importante ordem de insetos aquáticos, que possuem imaturos utilizados em programas de biomonitoramento da água. No Brasil, são encontradas 168 espécies, sendo 40 dessas ocorrentes no Estado do Rio de Janeiro. A taxonomia desta ordem atualmente é baseada na morfologia da armadura peniana dos machos, coloração e tamanho, mas muitas das descrições taxonômicas são incompletas, faltando assim associações entre machos, fêmeas e ninfas. O método mais utilizado para associação é criação dos imaturos para a obtenção dos adultos, entretanto, este é dificultado pela sensibilidade das ninfas à remoção do ambiente natural. Uma forma alternativa de associação é a utilização de ferramentas moleculares, sendo a utilização do marcador molecular COI utilizada com sucesso para a identificação em nível de espécie. No presente estudo, sequências de COI foram obtidas de adultos e ninfas de Plecoptera coletados em 106 pontos ao longo da Bacia do Rio Macaé (BRM). Adultos foram coletados com uma armadilha de luz do tipo Pensilvânia, enquanto as ninfas foram coletadas manualmente. O Rio Macaé é o maior rio que nasce no Estado do Rio de Janeiro e localiza-se na região norte do estado. O material foi identificado com uso de chaves específicas e descrições, e fêmeas e ninfas não identificadas foram morfotipadas. Dois exemplares de cada espécie/morfotipo foram separados para a análise molecular e imagens de microscopia eletrônica de varredura (MEV) que foi realizada a fim de buscar características para distinguir os espécimes. Um total de 1.248 espécimes foi coletado, sendo 132 machos, 266 fêmeas e 850 ninfas. Deste total, 44 táxons foram encontrados, distribuídos em 35 espécies, nove morfotipos não associados e cinco espécies novas. Dez ninfas são descritas e novos caracteres obtidos pela MEV foram adicionados a cinco espécies já descritas anteriormente; o macho de Kempnyia varipes é redescrito e a genitália desta espécie é ilustrada pela primeira vez. Trinta e um novos registros são feitos para a BRM e, 16 novos registros para o RJ. Foram sequenciados 134 espécimes de Plecóptera pertencentes a 40 espécies e 6 morfotipos, sendo 27 espécimes pertencentes à BRM e os demais de regiões do Sul e Sudeste do Brasil. As sequencias foram alinhadas no programa ClustalX, sendo as divergências genéticas par-a-par calculadas, ajustadas pelo modelo de evolução Kimura-2- parameter e uma análise de agrupamento de neighbor-joining (NJ) realizada no programa MEGA. As três árvores resultantes das análises de NJ realizadas em separado para Gripopterygidae, Anacroneuria e Kempnyia recuperaram um total de 46 linhagens genéticas distintas suportados também pela análise morfológica, sendo 27 de espécies da BRM. Com base nessas linhagens estabelecidas, as divergências interespecíficas variaram 3,3 a 25,3%, sendo as menores encontradas para Gripopteryx e as maiores para Anacroneuria. As divergências intraespecíficas variaram de zero a 15,1%, sendo as menores encontradas entre muitos espécimes da BRM e as maiores entre espécies de Kempnyia colossica (Navás).Os altos valores encontrados pra as espécies de plecoptera na BRM podem ser associados ao isolamento de espécies em topos de montanhas, ao isolamento geográfico ou mesmo se tratar de espécies crípticas. dc.description.abstractOther | Plecoptera is an important order of aquatic insects, and its immatures are used in water biomonitoring programs. In Brazil, there are 168 species recorded, 40 occurring in Rio de Janeiro State. The taxonomy of this order is based on male penial armature morphology, color, and measurements, but many taxonomic descriptions are incomplete, lacking male, female, and nymphal associations. The most commonly used method for immature-imago association is immature rearing to adulthood, however, this is hampered by the sensitivity of the nymphs to the removal from natural environment. An alternative form to immature-imago association is the use of molecular tools. The molecular marker COI have been used with success to species level identification. In this study, COI sequences were obtained from adults and nymphs collected in 106 localities in Macaé River Basin. Adults were collected with light traps and nymphs by manual method. All material were identified with specific keys and descriptions, and unidentified females and nymphs were morphotyped according the morphological characteristics. Two specimens from each morphotype were used in molecular analyze and to obtained images by scanning electronic microscope (SEM). SEM images were used to search for new characteristics to distinguish species. A total of 1248 specimens were collected, being 132 males, 266 females, and 850 nymphs. Out of this total, 44 taxa were found, distributed in 35 species, nine morphotypes not associated and five new species. Ten nymphs are described and new characters obtained by SEM were added to five known nymphs; the male of Kempnyia varipes was redescribed and the genitalia was illustrated for the first time. Thirty-one new records were made to Macaé River Basin and 16 characterized new records to RJ. Molecular material of 134 specimens of 40 species and six morphotypes were sequenced, being 27 species/morphotypes from Macaé River Basin and the others from Southern and Southeastern Brazil. The sequences were aligned with ClustalX software, being the pairwise divergences computed, adjusted with the Kimura 2-parameter model, and a cluster based on Neighbor Joining method (NJ) performed in MEGA software. Three resulting NJ trees are given for Gripopterygidae, Anacroneuria, and Kempnyia and recovered a total of 46 distinct genetic lineages also supported by the morphological analysis, where 27 were from MRB. Based in these lineages, the interespecific divergences ranged from 3.3 to 25.3%, with the lowest for Gripopteryx and higher for Anacroneuria. Intraspecific divergences ranged from zero to 15.1%, with the lowest found among many species from BRM and the highest among Kempnyia colossica (Navás) species. High values found for Plecoptera species from MRB could be associated to individuals being isolated on mountaintops, to geographical range, or to cryptic species.
