dc.description.abstractO interior das plantas abriga um grande número de bactérias diazotróficas endofíticas, que quando comparadas às diazotróficas exclusivamente rizosféricas, exibem grande contribuição para a nutrição das plantas hospedeiras via fixação biológica de Nitrogênio, provavelmente devido à proteção do complexo enzimático nitrogenase contra o oxigênio atmosférico, chegando a contribuir com até um terço do N acumulado em alguns genótipos de cana-deaçúcar. Experimentos conduzidos com inoculação isolada, combinada e conjunta de bactérias diazotróficas endofíticas das espécies Gluconacetobacter diazotrophicus, Azospirillum amazonense, Herbaspirillum seropedicae, Herbaspirillum rubrisubalbicans e Burkholderia tropica, demonstraram melhoras no desenvolvimento de plantas de cana contribuindo com até 31,4% do N acumulado em duas variedades de plantas cultivadas em condições ideais de solo e manejo cultural. Estes e outros estudos levaram à produção de um inoculante para a cultura de cana-de-açúcar por parte da Embrapa Agrobiologia, composto por cinco espécies de bactérias diazotróficas endofíticas testadas experimentalmente. Com a finalidade de localizar e caracterizar os sítios de colonização das estirpes bacterianas presentes neste inoculante para cana-de-açúcar, inoculadas isoladamente em toletes e plântulas de cana-de-açúcar, procedeuse a marcação das estirpes presentes no inoculante com o sistema de marcação repórter GFP, para a observação da locação física das bactérias em associação com a planta, através da visualização direta da proteína verde fluorescente (GFP), resultado alcançado com as estirpes HRC54 e HCC103 de H. rubrisubalbicans e H. seropedicae, respectivamente. Experimentos com Herbaspirillum rubrisubalbicans HCC103 e Herbaspirillum seropedicae HRC54 marcadas com GFP demonstraram que em ensaios de ascensão xilemática por fluxo transpiratório, HCC103GFP estava melhor distribuída no ambiente xilemático do que HRC54GFP. Em experimento piloto com plantas de arroz desinfestadas o padrão de colonização radicular superficial foi basicamente o mesmo, porém em mini-toletes e cariopses germinados há clara diferença no padrão espacial de colonização entre as estirpes. Herbaspirillum seropedicae HRC54GFP colonizou externamente gemas de mini-toletes inoculados e seus espaços intercelulares parenquimáticos na região do entrenó, enquanto que Herbaspirillum rubrisubalbicans HCC103GFP penetrou na gema e permaneceu predominantemente na região do nó, associada às paredes externas dos vasos xilemáticos. Os resultados sugerem que ocorre colonização diferencial tanto do sistema radicular quanto de colmos de cana-de-açúcar variedade RB867515 pelas estirpes HRC54 e HCC103 de H. rubrisubalbicans e H. seropedicae, respectivamente, sendo necessários estudos adicionais para melhor compreensão dos fatores que levam a esta colonização diferencial.por

dc.description.abstractOtherInside of plants lives a large number of diazotrophic bacteria, which when compared to exclusively rhizospheric diazotrophic exhibit great contribution to the nutrition of host plants through biological nitrogen fixation, probably due to the protection of the nitrogenase enzyme complex against atmospheric oxygen coming to contribute up to a third of N accumulated in some genotypes of sugarcane. Experiments with inoculum isolated, combined and joint diazotrophic bacteria species Gluconacetobacter diazotrophicus , Azospirillum amazonense Herbaspirillum seropedicae Herbaspirillum rubrisubalbicans, and Burkholderia tropica demonstrated improvements in the development of sugarcane plants contributing up to 31.4 % of the N accumulated in two varieties of plants grown in ideal conditions of soil and crop management. These and other studies led to the production of an inoculant for the culture of sugarcane by the Embrapa Agrobiologia, composed of five species of diazotrophic bacteria tested experimentally. In order to locate and characterize the colonization sites of the bacterial strains present in this inoculant to sugarcane, separately inoculated seedlings and stalks in sugarcane, proceeded to the marking of inoculant strains present in the system marking GFP reporter for the observation of physical location of bacteria in association with the plant, through the direct visualization of green fluorescent protein (GFP), results obtained with strains of H. seropedicae HRC54 and H. rubrisubalbicans HCC103. Experiments with Herbaspirillum rubrisubalbicans HCC103 and Herbaspirillum seropedicae HRC54 marked with GFP showed that in trials of ascent xylem by transpiration stream, HCC103GFP was better distributed environment in which the xylem HRC54GFP . In a pilot experiment with rice plants disinfected the surface root colonization pattern was basically the same on a ministalks and sprouted kernels no clear difference in the spatial pattern of colonization between strains. Herbaspirillum seropedicae HRC54GFP colonized externally buds of mini-stalks inoculated and their intercellular spaces in the parenchyma region of the internode, while Herbaspirillum rubrisubalbicans HCC103GFP penetrated the bud and remained predominantly in the node region, associated with the external walls of xilem vessels. The results suggest that differential colonization occurs much root system as the stalks of cane sugar variety RB867515 HRC54 and HCC103 by strains of H. rubrisubalbicans and H. seropedicae, respectively, and additional studies to better understand the factors that lead to this differential colonization .eng
