dc.description.abstractEsta tese descreve estudo fitoquímico e avaliação de algumas atividades biológicas de duas espécies neotropicais: Galianthe matogrossiana E.L. Cabral (Rubiaceae), Crateva tapia L. (Capparaceae) e detalha a definição da estrutura molecular de um novo alcaloide isolado de Justicia wasshauseniana Profice (Acanthaceae). Além da descrição dos procedimentos experimentais de isolamento descreve-se a identificação de estruturas moleculares de metabólitos especiais das plantas. Utilizaram-se técnicas cromatográficas e análise detalhada de RMN e EM neste trabalho. Fizeram-se avaliações de atividades biológicas como antinociceptiva, antiinflamatória e antioxidante. Identificaram-se 21 compostos nas frações apolares de raízes e partes aéreas de G. matogrossiana entre eles ácido graxo, antraquinona, derivado de chalcona e cumarina, derivados de ácido benzóico, esteroides e alguns triterpenos pentacíclicos. Nos ensaios in vivo (contorções abdominais, formalina e campo aberto em camundongos Swiss), com exceção da fração hexânica da raiz, as demais frações revelaram forte atividade antinociceptiva e antiinflamatória, inclusive com efeito superior a morfina. A atividade antioxidante, feita in vitro (cepas selvagens e Δyap1 de Saccharomyces cerevisiae), revelou que as frações hexânica e diclorometano possuem potentes ações quelantes de radicais. O ácido 3β-O-acetil-oleanólico inibiu amastigotas de Trypanosoma cruzi (IC50 = 11,06 μM; seletividade = 3,11). Estudos preliminares de docking molecular sugeriram afinidade promissora de triterpenos contra Aurora B quinase para aplicações anticâncer. Sobre C. tapia avaliou-se atividade antiglicante in vitro do extrato bruto de cascas, que não foi muito significativa. Até o momento isolaram-se da fração diclorometano o sitosterol glicosilado e lupeol. A discussão detalhada dos dados espectrométricos do alcaloide, isolado previamente de J. wasshauseniana, incluindo cálculos TD-DFT, definiu sua estrutura molecular incluindo a estereoqímica absoluta e foi nomeado como “brazoide E”. dc.description.abstractOtherThis thesis describes the phytochemical study and evaluation of some biological activities of two neotropical species: Galianthe matogrossiana E.L. Cabral (Rubiaceae) and Crateva tapia L. (Capparaceae), and details the definition of the molecular structure of a new alkaloid isolated from Justicia wasshauseniana Profice (Acanthaceae). In addition to the description of experimental isolation procedures, the identification of molecular structures of the plants’specialized metabolites is presented.Chromatographic techniques and detailed NMR and MS analyses were employed in this work. Biological activities such as antinociceptive, anti-inflammatory, and antioxidant effects were evaluated. Twenty-one compounds were identified in the nonpolar fractions ofroots and aerial parts of G. matogrossiana, including fatty acid, anthraquinone, chalcone and coumarin derivatives, benzoic acid derivatives, steroids, and some pentacyclic triterpenes. In vivo assays (abdominal writhing, formalin, and open-field tests in Swiss mice), except for the hexane root fraction, the other fractions showed strong antinociceptive and anti-inflammatory activity, even with effectssuperior to morphine. The in vitro antioxidant activity (wild-type and Δyap1 Saccharomyces cerevisiae strains) revealed that the hexane and dichloromethane fractions exhibited potent radical-chelating actions. 3-O-acetyl oleanolic acid inhibited Trypanosoma cruzi amastigotes (IC50 = 11.06 μM; selectivity = 3.11). Preliminary molecular docking studies suggested promising affinity of triterpenes toward Aurora B kinase for potential anticancer applications. Regarding C. tapia, in vitro antiglycation activity of the crude bark extract was evaluated and found to be not very significant. So far, β- sitosterol glycoside and lupeol have been isolated from the dichloromethane fraction. A detailed discussion of the spectrometric data of the alkaloid, previously isolated from J. wasshauseniana, including TD-DFT calculations, defined its molecular structure, including absolute stereochemistry, and it was named "brazoid E".
