| dc.description.abstract | O estresse oxidativo, resultante da produção excessiva de espécies reativas de oxigênio (EROs), provoca danos a biomoléculas e compromete processos celulares essenciais. Nesse contexto, a busca por substâncias antioxidantes naturais tem crescido, especialmente entre plantas alimentícias não convencionais (PANCs), como Xanthosoma sagittifolium (taioba), reconhecida pela presença de diversos metabólitos bioativos. No entanto, ainda são limitados os estudos que avaliam seus efeitos em modelos eucarióticos. Assim, este trabalho investigou o potencial antioxidante de extratos de folhas, caules e cormos de X. sagittifolium, bem como sua interação com o sistema antioxidante endógeno de Saccharomyces cerevisiae, utilizando as cepas BY4741 (selvagem) e Δyap1 (deficiente no fator de transcrição antioxidante Yap1p). Foram preparados extratos aquosos das partes aéreas por decocção, seguidos de particionamento com acetato de etila, além de extratos metanólico e diclorometânico dos cormos obtidos por extração a frio. A triagem fitoquímica por cromatografia em camada delgada revelou diferenças qualitativas entre os órgãos vegetais, com maior diversidade de compostos polares e semipolares nas folhas. A caracterização química por CLAE-DAD, análises espectrofotométricas e RMN confirmou a presença de ácidos fenólicos, flavonas e flavonoides C-glicosilados, como vitexina, isovitexina e orientina, além de metabólitos triterpênicos e esteroidais, especialmente nos extratos dos cormos. Os ensaios biológicos demonstraram ausência de citotoxicidade até a concentração de 50 mg/mL, utilizada como limite superior de triagem, indicando um perfil de segurança relevante para os extratos brutos. Embora relativamente elevada quando comparada a compostos isolados, essa concentração mostrou-se adequada para a avaliação inicial de misturas complexas, nas quais a atividade biológica decorre da ação combinada de múltiplos metabólitos. Na análise da oxidação intracelular, observou-se redução significativa dos níveis de EROs em ambas as cepas, com destaque para o extrato diclorometânico dos cormos, que reverteu completamente a oxidação induzida por peróxido de hidrogênio na linhagem Δyap1. Os testes de viabilidade celular evidenciaram efeito citoprotetor consistente, mais acentuado na cepa mutante. Adicionalmente, a análise funcional mitocondrial indicou recuperação do crescimento em meio não fermentativo e restabelecimento do potencial de membrana mitocondrial nas células Δyap1 tratadas, mesmo sob estresse oxidativo. Em conjunto, os resultados demonstram que Xanthosoma sagittifolium constitui uma fonte promissora de compostos antioxidantes naturais, capazes de modular o equilíbrio redox celular, preservar a integridade mitocondrial e compensar deficiências no sistema antioxidante endógeno, inclusive de forma independente da via regulada por Yap1p. Esses achados evidenciam não apenas a eficácia biológica dos extratos, mas também orientam a necessidade de estudos futuros de fracionamento e isolamento dos compostos bioativos, visando maior potência e aplicação nutracêutica, farmacológica e biotecnológica. | pt_BR |

| dc.description.abstractOther | Oxidative stress, resulting from the excessive production of reactive oxygen species (ROS), causes damage to biomolecules and compromises essential cellular processes. In this context, the search for natural antioxidant substances has increased, particularly among non-conventional food plants (PANCs), such as Xanthosoma sagittifolium (taro), which is recognized for its rich content of bioactive metabolites. However, studies evaluating its effects in eukaryotic models remain limited. Thus, this study investigated the antioxidant potential of extracts obtained from the leaves, stems, and corms of X. sagittifolium, as well as their interaction with the endogenous antioxidant system of Saccharomyces cerevisiae, using the BY4741 (wild-type) and Δyap1 (deficient in the antioxidant transcription factor Yap1p) strains. Aqueous extracts from the aerial parts were prepared by decoction, followed by partitioning with ethyl acetate, while methanolic and dichloromethane extracts from the corms were obtained through cold extraction. Phytochemical screening by thin-layer chromatography revealed qualitative differences among plant organs, with a greater diversity of polar and semi-polar compounds in the leaves. Chemical characterization by HPLC-DAD, spectrophotometric analyses, and NMR confirmed the presence of phenolic acids, flavones, and C-glycosylated flavonoids, such as vitexin, isovitexin, and orientin, as well as triterpenoid and steroidal metabolites, particularly in the corm extracts. Biological assays demonstrated no cytotoxicity up to a concentration of 50 mg/mL, used as the upper limit for screening, indicating a relevant safety profile for the crude extracts. Although relatively high compared to isolated compounds, this concentration proved suitable for the initial evaluation of complex mixtures, in which biological activity results from the combined action of multiple metabolites. Intracellular oxidation analysis showed a significant reduction in ROS levels in both strains, with particular emphasis on the dichloromethane corm extract, which completely reversed hydrogen peroxide-induced oxidation in the Δyap1 strain. Cell viability assays revealed a consistent cytoprotective effect, more pronounced in the mutant strain. Additionally, mitochondrial functional analysis indicated recovery of growth in non-fermentative medium and restoration of mitochondrial membrane potential in Δyap1 cells treated with the extracts, even under oxidative stress conditions. Taken together, the results demonstrate that Xanthosoma sagittifolium represents a promising source of natural antioxidant compounds capable of modulating cellular redox balance, preserving mitochondrial integrity, and compensating for deficiencies in the endogenous antioxidant system, including in a manner independent of the Yap1p-regulated pathway. These findings not only highlight the biological efficacy of the extracts but also support the need for future studies focused on fractionation and isolation of bioactive compounds, aiming to enhance potency and enable nutraceutical, pharmacological, and biotechnological applications. | en |
