The first aim of the present study was to evaluate the influence of estradiol treatment on female Wistar rats submitted to sodium overload model. For the first purpose, female Wistar rats with approximately 60 days old were ovariectomized (OVX), and treated with estradiol benzoate (EB-OVX) or vehicle (OVX) over 14 consecutive days and, by the end of this period, the female rats were submitted to a free-choice paradigm, with free access to water and hypotonic, isotonic or hypertonic saline solution. When submitted to free-choice paradigm with water and 0.3 M NaCl, both OVX and EB-OVX groups exhibited a reduced sodium preference, and although we did not observe estrogenic modulation in this case, the EB-OVX group presented a reduced preference for 0.15M NaCl, when compared to isotonic saline. In the saline overload model (where OVX and EB-OVX group were submitted to normal sodium diet and 0.3 NaCl as only fluid) we observed that estradiol inhibited the hypernatremia induced by this protocol, when compared to the respective OVX group, without modulating natriuresis. Although estradiol per se reduced systolic blood pressure and heart rate at baseline, the steroid modulated only partially the estradiol-mediated heart rate reduction. Finally, we observed that estradiol attenuated and reversed the reduction in plasma concentrations of ANG II and ANP, respectively, after 15 days of saline overload when compared to the respective control group. The second goal of this study was to investigate the influence of low sodium diet on rats submitted to acute sodium depletion (FuroCap) and β- adrenergic stimulation (Isoproterenol). For this, male Wistar rats with approximately 75 days of age were conditioned in a metabolic cage for adaptation for 3 consecutive days, with normal feed, water and hypertonic saline solution ad libitum. At the end of the adaptation period, the animals were randomly divided to receive low or normal sodium diet during 4 consecutive days with ad libitum access to water. The increase in fluid intake induced by FuroCap was exacerbated by the hyposodic diet, when compared to the respective control. Regarding the cardiovascular parameters, we observed that the hyposodic diet potentiated hypotension and tachycardia induced by acute sodium depletion. Furthermore, the low sodium diet attenuated the increase in plasma concentrations of oxytocin and angiotensin II while exacerbating the reduction in plasma ANP concentrations. Regarding β-adrenergic stimulation, the low sodium diet also exacerbated the increase in hypertonic water and saline intake induced by Isoproterenol when compared to the respective control. In contrast to the acute sodium depletion protocol, the hyposodic diet did not modulate Isoproterenol-induced hypotension and reflex bradycardia. Furthermore, it attenuated the increase in angiotensin II and atrial natriuretic peptide concentrations induced by Isoproterenol. Taken together, we can conclude that: i) estradiol seems to exert an attenuating effect on the deleterious consequences of a high sodium diet; and ii) the low sodium diet potentiates ingestive behaviors induced by acute sodium depletion (FuroCap) and β-adrenergic stimulation (Isoproterenol) but attenuates neuroendocrine factors influenced by these protocols.en
