dc.description.abstract | A dificuldade no diagnóstico da paratuberculose na fase assintomática está relacionada principalmente à resposta imune do hospedeiro frente aos diversos estágios da enfermidade; a maioria dos animais infectados por Mycobacterium avium subsp. paratuberculosis (Map) não apresenta e não apresentará sinais clínicos característicos da enfermidade ao longo de sua vida e pode eliminar, intermitentemente, o agente infeccioso no ambiente, através das fezes, mantendo a infecção no rebanho. Atualmente, os testes laboratoriais disponíveis para diagnóstico subclínico da paratuberculose apresentam baixa eficiência, o que limita o diagnóstico precoce da enfermidade e contribui para a ineficiência das medidas de controle da infecção no rebanho. O desenvolvimento de técnicas confiáveis e específicas para a detecção “post-mortem” da paratuberculose possibilita o diagnóstico antes do aparecimento dos sinais clínicos, e pode auxiliar na elaboração de novos testes para detecção “in vivo” da infecção. No Brasil, a paratuberculose já foi descrita em diversas espécies de ruminantes e em vários estados, no entanto, não há programas sanitários oficiais para o controle da enfermidade. Um dos objetivos deste trabalho foi detectar a infecção pré-clínica da paratuberculose em bovinos oriundos de rebanho positivo para paratuberculose por metodologias de diagnóstico anátomopatológico. Foram realizados estudos histopatológico, histoquímico e imuno-histoquímico de intestino delgado e linfonodos mesentéricos coletados no frigorífico local. À histopatologia, observaram-se lesões compatíveis com paratuberculose em animais assintomáticos; bacilos álcool-ácido resistentes não foram visualizados e a imuno-histoquímica mostrou marcação positiva em alguns fragmentos analisados. A partir dos resultados, podemos afirmar que o teste de imuno-histoquímica pode ser uma importante ferramenta para detecção de Map, especialmente em tecidos com lesões tuberculoide ou “paucibacillary”, enriquecendo os métodos convencionais de diagnóstico histopatológico para confirmação da infecção. Além disso, em situações onde não há disponibilidade de material para cultivo bacteriano pode-se utilizar desta metodologia para conclusão do diagnóstico. Na segunda parte do trabalho, foi realizado um estudo comparativo de duas soluções fixadoras de tecido, uma contendo formol, substância tradicionalmente utilizada em laboratórios de patologia e a outra composta por sais de zinco. O intuito do estudo foi avaliar a manutenção da histomorfologia, a capacidade de detecção de bacilos álcool-ácido resistentes e antígenos de Mycobacterium spp. por imunohistoquímica e amplificação dos fragmentos de DNA bovino e de Mycobacterium avium subsp. paratuberculosis por PCR GAPDH, IS900 e F57, em fragmentos de íleo e linfonodo mesentérico fixados, de bovino com paratuberculose clínica. Avaliaram-se, também, possíveis alterações causadas em diferentes períodos de tempo em que os fragmentos foram mantidos na solução fixadora. Verificou-se notável manutenção da arquitetura do tecido em fragmentos fixados em solução de zinco, com qualidade da imagem histológica similar aos tecidos fixados em formol; os resultados obtidos pela coloração de Ziehl-Neelsen e imunohistoquímica foram semelhantes nos dois fixadores. Já a PCR mostrou amplificação apenas nas amostras fixadas em zinco, o que demonstra que este fixador pode ser uma ferramenta importante para manutenção de ácidos nucleicos e macromoléculas para estudos genéticos e proteômicos. | por |

dc.description.abstractOther | The difficulty of diagnosis of subclinical paratuberculosis (Johne´s disease), caused by Mycobacterium avium subsp. paratuberculosis (Map), is mainly related to host immune responses to the different stages of infection; the majority of infected animals does not and will not present throughout their life clinical signs characteristic of the disease, and can intermittently eliminate the infectious agent through feces into the environment, keeping the infection in the herd. Currently, the laboratory tests available for the diagnosis of subclinical paratuberculosis are unsatisfactory, limiting the early diagnostic of the disease and contribute to the inefficiency of control measures for infection in the herd. The development of reliable and specific techniques for detecting paratuberculosis at “post-mortem” examination could allow preclinical diagnosis contribute to development of a new test for “in vivo” diagnosis of the infection. In Brazil, paratuberculosis has been reported from different ruminant species and has a broad geographic distribution; however, there exists no official program for control of the disease. An objective of this study was to detect preclinical infection of paratuberculosis in cattle through pathological methodologies for the diagnosis in paratuberculosis positive herds. Histopathological, histochemical and immunohistochemical studies of the small intestine and mesenteric lymph nodes collected in the slaughter house were performed. Histopathology revealed lesions compatible with paratuberculosis in asymptomatic cattle, without visualization of acid-fast bacilli, and immunohistochemistry resulted in positive staining in some fragments analyzed. The results obtained in this study show that immunohistochemistry could be an important tool for detecting Map, especially in tissues with few mycobacteria, and enrich conventional methods for histopathological confirmation of the diagnosis of Johne´s disease. Additionally, in situations where there is no availability of material for culture, this methodology could be used for diagnostic conclusion. - In the second part of this study, we compared two tissue fixative solutions: one containing formaldehyde, traditionally used in pathology laboratories, and the other consisting of zinc salts. The purpose was to evaluate (1) the maintenance of histomorphology, (2) the ability to detect acid-fast bacilli and antigens of Mycobacterium spp. by immunohistochemistry, and (3) the amplification of DNA fragments of Mycobacterium avium subsp. paratuberculosis by PCR IS900 and F57 in sections of formaldehyde fixed ileum and mesenteric lymph nodes from cattle with paratuberculosis. Also were evaluated possible variations caused by different time periods during which the fragments were kept in the fixative solution. We found that (1) the zinc-based fixative showed remarkable maintenance of tissue architecture with similar histological image quality of tissues fixed in formalin, (2) the results obtained by Ziehl- Neelsen staining and immunohistochemistry were similar with the two fixative solutions used. However, PCR amplification was observed only with samples fixed in zinc salts, what demonstrates that this latter fixative can be an important tool for maintenance of nucleic acids and macromolecules for genetic and proteomic studies. | eng |
